Abstract

This invention relates to novel compounds having the formula (I): and to their pharmaceutical compositions and to their methods of use. These novel compounds provide a treatment for cancer.

The invention claimed is: 
     
       1. A compound of formula (I): 
       
         
           
           
               
               
           
         
         wherein: 
         R 1  and R 2  are independently selected from hydrogen, halo, nitro, cyano, hydroxy, trifluoromethoxy, amino, carboxy, carbamoyl, mercapto, sulphamoyl, C 1-6 alkyl, C 2-6 alkenyl, 
         C 1-6 alkoxy, C 1-6 alkanoyl, C 1-6 alkanoyloxy, N—(C 1-6 alkyl)amino, N,N-(C 1-6 alkyl) 2 -amino, 
         C 1-6 alkanoylamino, N—(C 1-6 alkyl)carbamoyl, N,N-(C 1-6 alkyl) 2 -carbamoyl, C 1-6 alkylS(O) a  wherein a is 0 to 2, C 1-6 alkoxycarbonyl, N—(C 1-6 alkyl)sulphamoyl, N,N-(C 1-6 alkyl) 2 sulphamoyl C 1-6 alkylsulphonylamino and cyclopropyl, 
         R 4  is C 1-6 alkyl, which may be optionally substituted on carbon by one or more hydroxy groups; 
         R 34  is H or C 1-6 alkyl, which may be optionally substituted on carbon by one or more hydroxy groups; 
         Ring C is phenyl, pyridyl, or pyrimidinyl; 
         R 5  is independently selected from halo, nitro, cyano, hydroxy, trifluoromethoxy, amino, carboxy, carbamoyl, mercapto, sulphamoyl, C 1-6 alkyl, C 2-6 alkenyl, C 7-6 alkynyl, C 1-6 alkoxy, C 1-6 alkanoyl, C 1-6 alkanoyloxy, N—(C 1-6 alkyl)amino, N,N—(C 1-6 alkyl) 2 -amino, C 1-6 alkanoylamino, N—(C 1-6 alkyl)carbamoyl, N,N—(C 1-6 alkyl) 2 -carbamoyl, C 1-6 alkylS(O), wherein a is 0 to 2, C 1-6 alkoxycarbonyl, N—(C 1-6 alkyl)sulphamoyl, N,N—(C 1-6 —alkyl) 2 sulphamoyl and C 1-6 alkylsulphonylamino; 
         n is 0, 1, 2 or 3; 
         R 8 , R 9  and R 10  are independently selected from hydrogen, halo, nitro, cyano, hydroxy, trifluoromethoxy, amino, carboxy, carbamoyl, mercapto, sulphamoyl, and C 1-6 alkyl; 
         or a pharmaceutically acceptable salt thereof. 
       
     
     
       2. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 1  is isopropoxy. 
     
     
       3. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 1  is cyclopropyl. 
     
     
       4. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 1  is t-butyl. 
     
     
       5. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 2  is hydrogen. 
     
     
       6. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 4  is methyl. 
     
     
       7. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 34  is hydrogen. 
     
     
       8. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein Ring C is pyridyl. 
     
     
       9. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein Ring C is pyrid-2-yl. 
     
     
       10. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein Ring C is pyrid-3-yl. 
     
     
       11. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 5  is halo. 
     
     
       12. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein R 5  is fluorine. 
     
     
       13. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , wherein n is 1 or 2. 
     
     
       14. A compound of formula (I) or a pharmaceutically acceptable salt thereof, as claimed in  claim 13 , wherein n is 1. 
     
     
       15. A compound selected from:
 (S)-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-fluoro-2-(1-(4-fluorophenyl)ethylamino)-4-(methylamino)nicotinonitrile; 
 (S)-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-fluoro-2-(1-(5-fluoropyridin-2-yl)ethylamino)nicotinonitrile; 
 (S)-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-fluoro-2-(1-(4-fluorophenyl)ethylamino)-4-(isopropylamino)nicotinonitrile; 
 (S)-5-chloro-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-2-(1-(5-fluoropyridin-2-yl)ethylamino)nicotinonitrile; 
 6-[(5-cyclopropyl-1H-pyrazol-3-yl)amino]-2-{[(1S)-1-(3,5-difluoropyridin-2-yl)ethyl]amino}-5-fluoronicotinonitrile; 
 (S)-5-fluoro-2-(1-(4-fluorophenyl)ethylamino)-6-(5-isopropoxy-1H-pyrazol-3-ylamino)nicotinonitrile; 
 (S)-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-fluoro-2-(1-(4-fluorophenyl)ethylamino)nicotinonitrile; 
 (R)-6-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-fluoro-2-(1-(4-fluorophenyl)-2-hydroxyethylamino)nicotinonitrile; and 
 (R)-5-fluoro-2-(1-(4-fluorophenyl)-2-hydroxyethylamino)-6-(5-isopropoxy-1H-pyrazol-3-ylamino)nicotinonitrile; 
 or a pharmaceutically acceptable salt thereof. 
 
     
     
       16. A pharmaceutical composition comprising a compound of formula (I), or a pharmaceutically acceptable salt thereof, as claimed in  claim 1 , together with at least one pharmaceutically acceptable carrier, diluent or excipient. 
     
     
       17. A pharmaceutical composition comprising a compound as claimed in  claim 15 , or a pharmaceutically acceptable salt thereof, together with at least one pharmaceutically acceptable carrier, diluent or excipient.
