Abstract
The present invention relates to novel human secreted proteins and isolated nucleic acids containing the coding regions of the genes encoding such proteins. Also provided are vectors, host cells, antibodies, and recombinant methods for producing human secreted proteins. The invention further relates to diagnostic and therapeutic methods useful for diagnosing and treating diseases, disorders, and/or conditions related to these novel human secreted proteins.
Claims
exact text as granted — not AI-modified1. An isolated antibody or fragment thereof that specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of amino acid residues 1 to 133 of SEQ ID NO:75;
(b) a protein consisting of amino acid residues 77 to 127 of SEQ ID NO:75;
(c) a protein consisting of a portion of SEQ ID NO:75, wherein said portion comprises at least 30 contiguous amino acid residues of SEQ ID NO:75; and
(d) a protein consisting of a portion of SEQ ID NO:75, wherein said portion comprises at least 50 contiguous amino acid residues of SEQ ID NO:75.
2. The antibody or fragment thereof of claim 1 that specifically binds protein (a).
3. The antibody or fragment thereof of claim 1 that specifically binds protein (b).
4. The antibody or fragment thereof of claim 1 that specifically binds protein (c).
5. The antibody or fragment thereof of claim 1 that specifically binds protein (d).
6. The antibody or fragment thereof of claim 2 that specifically binds protein (b).
7. The antibody or fragment thereof of claim 3 wherein said protein bound by said antibody or fragment thereof is glycosylated.
8. The antibody or fragment thereof of claim 3 which is a human antibody.
9. The antibody or fragment thereof of claim 3 which is a polyclonal antibody.
10. The antibody or fragment thereof of claim 3 which is selected from the group consisting of:
(a) a chimeric antibody;
(b) a humanized antibody;
(c) a single chain antibody; and
(d) a Fab fragment.
11. A labeled antibody or fragment thereof, wherein the, antibody or fragment thereof of claim 3 is labeled.
12. The labeled antibody or fragment thereof of claim 11 wherein the labeled is selected from the group consisting of:
(a) an enzyme;
(b) a fluorescent label;
(c) a luminescent label; and
(d) a bioluminescent label.
13. The antibody or fragment thereof of claim 3 wherein said antibody or fragment thereof specifically binds to said protein in a Western blot.
14. The antibody or fragment thereof of claim 3 wherein said antibody or fragment thereof specifically binds to said protein in an ELISA.
15. An isolated cell that produces the antibody or fragment thereof of claim 3 .
16. A hybridoma that produces the antibody of claim 3 .
17. A method of detecting a secreted protein HTEBY11 (SEQ ID NO: 75) in a biological sample comprising:
(a) contacting the biological sample with the antibody or fragment thereof of claim 3 ; and
(b) detecting the protein in the biological sample.
18. The method of claim 17 wherein the antibody or fragment thereof is a polyclonal antibody.
19. An isolated antibody obtained from an animal that has been immunized with a protein selected from the group consisting of:
(a) a protein comprising the amino acid sequence of amino acid residues 1 to 133 of SEQ ID NO:75;
(b) a protein consisting of the amino acid sequence of amino acid residues 77 to 127 of SEQ ID NO:75;
(c) a protein consisting of the amino acid sequence of at least 30 contiguous amino acid residues of SEQ ID NO:75; and
(d) a protein consisting of the amino acid sequence of at least 50 contiguous amino acid residues of SEQ ID NO:75;
wherein said antibody or fragment thereof specifically binds to said amino acid sequence.
20. The antibody of claim 19 obtained from an animal immunized with protein (a).
21. The antibody of claim 19 obtained from an animal immunized with protein (b).
22. The antibody of claim 19 obtained from an animal immunized with protein (c).
23. The antibody of claim 19 obtained from an animal immunized with protein (d).
24. The antibody of claim 19 which is a monoclonal antibody.
25. The antibody of claim 19 which is selected from the group consisting of:
(a) a chimeric antibody;
(b) a polyclonal antibody;
(c) a humanized antibody;
(d) a single chain antibody; and
(e) a Fab fragment.
26. An isolated monoclonal antibody or fragment thereof that specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of amino acid residues 1 to 133 of SEQ ID NO:75;
(b) a protein consisting of amino acid residues 77 to 127 of SEQ ID NO:75;
(c) a protein consisting of a portion of SEQ ID NO:75, wherein said portion comprises at least 30 contiguous amino acid residues of SEQ ID NO:75; and
(d) a protein consisting of a portion of SEQ ID NO:75, wherein said portion comprises at least 50 contiguous amino acid residues of SEQ ID NO:75.
27. The antibody or fragment thereof of claim 26 that specifically binds protein (a).
28. The antibody or fragment thereof of claim 26 that specifically binds protein (b).
29. The antibody or fragment thereof of claim 26 that specifically binds protein (c).
30. The antibody or fragment thereof of claim 26 that specifically binds protein (d).
31. The antibody or fragment thereof of claim 27 that specifically binds protein (b).
32. The antibody or fragment thereof of claim 28 wherein said protein bound by said antibody or fragment thereof is glycosylated.
33. The antibody or fragment thereof of claim 28 which is a human antibody.
34. The antibody or fragment thereof of claim 28 which is selected from the group consisting of:
(a) a chimetic antibody;
(b) a humanized antibody;
(c) a single chain antibody; and
(d) a Fab fragment.
35. A labeled antibody or fragment thereof, wherein the, antibody or fragment thereof of claim 28 is labeled.
36. The labeled antibody or fragment thereof of claim 35 wherein the label is selected from the group consisting of:
(a) an enzyme;
(b) a fluorescent label;
(c) a luminescent label; and
(d) a bioluminescent label.
37. The antibody or fragment thereof of claim 28 wherein said antibody or fragment thereof specifically binds to said protein in a Western blot.
38. The antibody or fragment thereof of claim 28 wherein said antibody or fragment thereof specifically binds to said protein in an ELISA.
39. An isolated cell that produces the antibody or fragment thereof of claim 28 .
40. A hybridoma that produces the antibody of claim 28 .
41. A method of detecting a secreted protein HTEBY11 in a biological sample comprising:
(a) contacting the biological sample with the antibody or fragment thereof of claim 28 ; and
(b) detecting the protein in the biological sample.
42. An isolated antibody or fragment thereof that specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of the full-length polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(b) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, comprising the amino acid sequence of amino acid residues 77 to 127 of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(c) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, wherein said portion comprises at least 30 contiguous amino acid residues of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070; and
(d) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, wherein said portion comprises at least 50 contiguous amino acid residues of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070.

(d) a bioluminescent label.
54. The antibody or fragment thereof of claim 44 wherein said antibody or fragment thereof specifically binds to said protein in a Western blot.
55. The antibody or fragment thereof of claim 44 wherein said antibody or fragment thereof specifically binds to said protein in an ELISA.
56. An isolated cell that produces the antibody or fragment thereof of claim 44 .
57. A hybridoma that produces the antibody of claim 44 .
58. A method of detecting a protein in a biological sample comprising:
(a) contacting the biological sample with the antibody or fragment thereof of claim 44 ; and
(b) detecting the protein in the biological sample.
59. The method of claim 58 wherein the antibody or fragment thereof is a polyclonal antibody.
60. An isolated antibody obtained from an animal that has been immunized with a protein selected from the group consisting of:
(a) a protein comprising the amino acid sequence of the full-length polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(b) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, comprising the amino acid sequence of amino acid residues 77 to 127 of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(c) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, comprising the amino acid sequence of at least 30 contiguous amino acid residues of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070; and
(d) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, comprising the amino acid sequence of at least 50 contiguous amino acid residues the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
wherein said antibody specifically binds to said amino acid sequence.

68. An isolated monoclonal antibody or fragment thereof at specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of the full-length polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(b) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Number 97922 or 209070, comprising the amino acid sequence of amino acid residues 77 to 127 of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070;
(c) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, wherein said portion comprises at least 30 contiguous amino acid residues of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070; and
(d) a protein consisting of a portion of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070, wherein said portion comprises at least 50 contiguous amino acid residues of the polypeptide encoded by the HTEBY11 cDNA contained in ATCC Deposit Numbers 97922 or 209070.

(a) contacting the biological sample with the antibody or fragment thereof of claim 70 ; and
(b) detecting the protein in the biological sample.

An isolated antibody or fragment thereof that specifically binds a secreted HTEBY11 (SEQ ID NO: 75) protein purified from a cell culture wherein the cells in said cell culture comprise a polynucleotide encoding amino acids 1 to 133 of SEQ ID NO:75 operably associated with a regulatory sequence that controls the expression of said polynucleotide.
85. The antibody or fragment thereof of claim 84 which is a monoclonal antibody.
86. The antibody or fragment thereof of claim 84 which is a human antibody.
87. The antibody or fragment thereof of claim 84 which is selected from the group consisting of:
(a) a chimeric antibody;
