Vaccines, the process for preparing the same and the applications thereof
Abstract
This invention relates to a process for preparing vaccines, which consists in bringing into contact a toxic product with glutaraldehyde at a concentration and for a duration just sufficient to detoxify or inactivate said toxic product, and in stopping the glutaraldehyde-antigen reaction as soon as said inactivation stage is reached by physical and/or chemical means. The thus obtained inactivate product remaining antigenic and retaining its immunizing power, is usable as a vaccine for the preventive treatment of human and animals against infections caused for example by microbial or venomous toxins.

1. A process for preparing a bacterial toxin comprising the steps of: a. reacting a bacterial toxin with glutaraldehyde of a concentration ranging from about 0.00131 M to about 0.0526 M and for a duration just sufficient for almost complete detoxification within 1 hour with complete detoxification within 3 hours, said detoxification being equivalent to formaldehyde detoxification requiring a period of not less than 2 weeks, to detoxify and inactivate said toxin, b. stopping the reaction as soon as said inactivation stage is reached, whereby an inactivated product is obtained which is usable as a bacterial toxin while remaining antigenic and retaining its immunizing power.
2. A process according to claim 1, wherein said toxin is diphtherial toxin.
3. A process according to claim 1, wherein said toxin is tetanus toxin.
4. A process according to claim 1, wherein α-staphylococcus toxin is used as the toxin.
5. A process according to claim 1, wherein β-staphylococcus toxin is used as the toxin.
6. A process according to claim 1, wherein the glutaraldehyde and said toxin are brought into contact at a temperature of from ambient temperature to about 40° C.
7. A process according to claim 6, wherein said temperature is between 35° and 40° C.
8. A process according to claim 7, wherein said temperature is about 37° C.
9. A process according to claim 1, wherein the reaction between said toxin and glutaraldehyde is stopped by the addition of an agent able to block the glutaraldehyde-antigen reaction or to react with the glutaraldehyde which is in the free state in the reaction medium and selected from the group consisting of amino-acids and inorganic salts.
10. A process according to claim 9, wherein said agent is an amino-acid.
11. A process according to claim 10, wherein said amino-acid is lysine or glycin.
12. A process according to claim 9, wherein said agent is sodium bisulfite.
13. A process according to claim 1, wherein said reaction is stopped by subjecting the detoxified product to dialysis against an aqueous solution of an inorganic salt, whereby a fluid product usable as a bacterial toxin is obtained.
14. A process according to claim 1, wherein said agent is an inorganic salt selected from the group consisting of sodium phosphate and sodium chloride.
15. A process according to claim 13, wherein merthiolate is added to said aqueous solution used for said dialysis.
16. A process according to claim 1, wherein said reaction is stopped by centrifuging and washing the detoxified product to remove from it the excess of glutaraldehyde.
17. A process according to claim 1, wherein a sterile caclium chloride solution is added to the detoxified product obtained in fluid form by dialysis against a buffer solution, whereby an adsorbed product useful as a bacterial toxin is obtained.
18. A process according to claim 13, wherein aluminum hydroxide is added to said detoxified bacterial toxin, obtained by dialysis against a sodium chloride solution, whereby an adsorbed product useful as a bacterial toxin is obtained.
19. A process according to claim 13, wherein aluminum phosphate is added to said detoxified bacterial toxin, obtained by dialysis against a sodium chloride solution, whereby an adsorbed product useful as a bacterial toxin is obtained.
20. A bacterial toxin obtained by the process according to claim 1 and containing detoxified derivatives corresponding to the toxic products of a bacterial toxin selected from the group consisting of diphtherial toxin, tetanus toxin, and α- and β-staphylococcus toxins.
21. A bacterial toxin according to claim 20, further containing an agent selected from the group consisting of amino-acids and inorganic salts.
22. A bacterial toxin according to claim 21, wherein said agent is an amino-acid.
23. A bacterial toxin according to claim 21, wherein said agent is selected from the group of lysine and glycin.
24. A bacterial toxin according to claim 20, in the fluid form.
25. A bacterial toxin according to claim 20, in the adsorbed form.
26. A bacterial toxin according to claim 22, adsorbed on calcium phosphate, aluminum hydroxide or aluminum phosphate.
