Abstract

The invention provides a compound of formula (I): or a salt thereof, wherein R 1 -R 2 and X and Y have any of the values described in the specification, as well as compositions comprising a compound of formula (I). The compounds are useful as contraceptive agents.

 R 1  is a 6-10 membered aryl, (C 3 -C 6 )cycloalkyl, or a 5-10 membered heteroaryl, which 6-10 membered aryl, (C 3 -C 6 )cycloalkyl, and 5-10 membered heteroaryl is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy;

 ring A is phenyl, 3-8 membered heterocyclic ring, or a 5-10 membered heteroaryl, which phenyl, 3-8 membered heterocyclic ring and 5-10 membered heteroaryl is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy; 
 R 2  is —C(═O)—NR a R b  or a 5-membered heteroaryl ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, NR c R d , and (C 2 -C 6 )alkanoyloxy; 
 R a  is H or (C 1 -C 6 )alkyl; and R b  is H, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, or (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, and (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, aryl, 5-10 membered heteroaryl, NR e R f , and (C 2 -C 6 )alkanoyloxy; or 
 R a  and R b  together with the nitrogen to which they are attached form a 3-8 membered heterocyclic ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, (C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy; 
 each R c  and R d  is independently selected from the group consisting of H, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, and (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, or R c  and R d  together with the nitrogen to which they are attached form a 3-8 membered heterocyclic ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, (C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy; and 
 each R e  and R f  is independently selected from the group consisting of H, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, and (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, wherein (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, and (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl are optionally substituted with one or more halo, or R e  and R f  together with the nitrogen to which they are attached form a 3-8 membered heterocyclic ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, (C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy. 
 
       
     
     
         2 . The compound of  claim 1  or a salt thereof, wherein:
 R 1  is a 6-10 membered aryl, (C 3 -C 6 )cycloalkyl, or a 5-10 membered heteroaryl, which 6-10 membered aryl, (C 3 -C 6 )cycloalkyl, and 5-10 membered heteroaryl is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy;

The compound or salt of  claim 1 , wherein ring A is phenyl that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy.

The compound or salt of  claim 1 , wherein ring A is a 3-8 membered heterocyclic ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy.

The compound or salt of  claim 1 , wherein ring A is a 5-10 membered heteroaryl that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy.

The compound or salt of  claim 1 , wherein R 1  is a (C 3 -C 6 )cycloalkyl that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy.

The compound or salt of  claim 1 , wherein R 1  is 4-methylphenyl, 3-methylphenyl, 2-methylphenyl, 4-fluorophenyl, 4-methoxyphenyl, 4-trifluoromethylphenyl, 4-chlorophenyl, 3,4-dichlorophenyl, cyclohexyl, naphthyl, 3-trifluoromethylphenyl, 3-methoxyphenyl, or 3-chlorophenyl. 
     
     
         11 . The compound or salt of  claim 1 , wherein X is N and Y is N or X is CH and Y is CH. 
     
     
         12 . The compound or salt of  claim 1 , wherein R 2  is —C(═O)—NR a R b .

The compound or salt of  claim 1 , wherein R 2  is a 5-membered heteroaryl ring that is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy, wherein any (C 1 -C 6 )alkyl, (C 3 -C 6 )cycloalkyl, (C 3 -C 6 )cycloalkyl(C 1 -C 6 )alkyl, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, and (C 2 -C 6 )alkanoyloxy is optionally substituted with one or more groups independently selected from the group consisting of halo, hydroxy, carboxy, nitro, (C 1 -C 6 )alkoxy, (C 1 -C 6 )alkanoyl, (C 1 -C 6 )alkoxycarbonyl, (C 1 -C 6 )alkylthio, NR c R d , and (C 2 -C 6 )alkanoyloxy.

         17 . A method to reduce the likelihood of conception following intercourse between a male subject and a female subject, comprising administering a compound or pharmaceutically acceptable salt as described in  claim 1  to the male subject or to the female subject. 
     
     
         18 . A method to reduce sperm motility in a male subject, comprising administering a compound or pharmaceutically acceptable salt as described in  claim 1  to the male subject. 
     
     
         19 . A method to reduce sperm motility following intercourse between a male subject and a female subject, comprising administering a compound or pharmaceutically acceptable salt as described in  claim 1  to the female subject. 
     
     
         20 . A method to produce reversible infertility in a male subject, comprising administering a compound or pharmaceutically acceptable salt as described in  claim 1  to the male subject.
