Ambrisentan for use in the treatment of acute renal failure

Abstract

Ambrisentan and formulations thereof for use in the treatment and prevention of acute renal failure associated with renal vasoconstriction.

1 . A method for pre-treating of a kidney comprising administering an effective amount of ambrisentan to the kidney, wherein ambrisentan is administered to the kidney such that the levels of ambrisentan in a vascular space of the kidney are maintained below about 1 ng/ml, to pre-treat the kidney. 
     
     
         2 . A method for preserving a kidney awaiting a transplant, comprising administering an effective amount of ambrisentan to the kidney, wherein ambrisentan is administered to the kidney such that the levels of ambrisentan in a vascular space of the kidney are maintained below about 1 ng/ml, to preserve the kidney awaiting the transplant. 
     
     
         3 . A liquid composition for intravenous administration comprising an ambrisentan, a buffer and a solvent. 
     
     
         4 . The composition  claim 3 , wherein the pH of the composition is at least 7. 
     
     
         5 . The composition according to  claim 3 , wherein the liquid composition is free from water. 
     
     
         6 . An ampoule containing the liquid composition according to  claim 3 , wherein the ambrisentan is present at a concentration of at least 0.001 wt % and at most 5 wt %, based on the total weight of the liquid composition. 
     
     
         7 . A pre-filled syringe containing the liquid composition according to  claim 3 , wherein the ambrisentan is present at a concentration of at least 0.001 wt % and at most 1 wt %, based on the total weight of the liquid composition. 
     
     
         8 . A cartridge for a subcutaneous pump or medication reservoir for a subcutaneous pump containing the liquid composition according to  claim 3 , wherein the ambrisentan is present at a concentration of at least 0.01 wt % and at most 1 wt %, based on the total weight of the liquid composition. 
     
     
         9 . A cartridge for a subcutaneous pen containing the liquid composition according to  claim 3 , wherein the ambrisentan is present at a concentration of at least 0.01 wt % and at most 1 wt %, based on the total weight of the liquid composition. 
     
     
         10 . An IV container containing the liquid composition according to  claim 3 , wherein the ambrisentan is present at a concentration of at least 0.00001 wt % and at most 0.05 wt %, based on the total weight of the liquid composition.
