Abstract

Provided herein are antibodies, or antigen binding portions thereof, that bind to glucocorticoid-inducible TNF receptor (GITR). Also provided are uses of these proteins in therapeutic applications, such as in the treatment of cancer. Further provided are cells that produce the antibodies, polynucleotides encoding the heavy and/or light chain variable region of the antibodies, and vectors comprising the polynucleotides encoding the heavy and/or light chain variable region of the antibodies.

         32 . The isolated antibody of  claim 29 , wherein the antibody binds to a cell expressing membrane-bound human GITR with a K D  of 10 nM or less as determined by Scatchard analysis. 
     
     
         33 . The isolated antibody of  claim 29 , wherein the antibody binds to human GITR on activated human T cells. 
     
     
         34 . The isolated antibody of  claim 29 , wherein the antibody stimulates a T cell response. 
     
     
         35 . The isolated antibody of  claim 29 , which is a human IgG antibody. 
     
     
         36 . The isolated antibody of  claim 35 , which is an IgG1, an IgG2, an IgG3, or an IgG4 antibody. 
     
     
         37 . The isolated antibody of  claim 29 , wherein the antibody is a human or humanized antibody. 
     
     
         38 . A bispecific molecule comprising the antibody of  claim 29  linked to a molecule having a second binding specificity. 
     
     
         39 . A nucleic acid encoding the heavy and/or light chain variable region of the antibody of  claim 29 . 
     
     
         40 . An expression vector comprising the nucleic acid of  claim 39 . 
     
     
         41 . A cell transformed with the expression vector of  claim 40 . 
     
     
         42 . An immunoconjugate comprising the antibody of  claim 29 , linked to an agent.

         45 . A method of preparing an antibody which binds to human GITR comprising culturing the cell of  claim 41  and isolating the antibody from the cell. 
     
     
         46 . A method for inhibiting the growth of tumor cells in a subject comprising administering to the subject an effective amount of the antibody of  claim 29 . 
     
     
         47 . A method of treating cancer comprising administering to a subject in need thereof a therapeutically effective amount of the antibody of  claim 29 . 
     
     
         48 . A method of detecting the presence of human GITR in a sample comprising contacting the sample with the antibody of  claim 29  under conditions that allow for formation of a complex between the antibody and human GITR, and detecting the formation of a complex.
