Abstract
The subject matter herein is directed to carbazole-containing amide, carbamate, and urea derivatives and pharmaceutically acceptable salts or hydrates thereof of structural formula I wherein the variable R 1 , R 2 , R 3 , R 4 , R 5 , R 6 , R 7 , A, D, E, G, J, L, M, Q, a, and b are accordingly described. Also provided are pharmaceutical compositions containing the compounds of formula I to treat a Cry-mediated disease or disorder, such as diabetes, complications associated with diabetes, Cushing's syndrome, NASH, NAFLD, asthma, and COPD.

c is 1 or 2;
d is 0, 1, or 2; and
e, f, and g are each independently 0, 1, 2, 3, 4, or 5.

The compound according to claim 1 , wherein each of A, D, E, G, J, L, M, and Q are C; each of R 1 and R 2 is independently selected from H or halo; R 4 is H or (C 1 -C 6 )alkyl, R 3 and R 5 are H; R 6 and R 7 are linked to each other as a 4-12 membered mono- or bicyclic amide ring; R 8 , R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , a, b, c, d, e, and f are as defined herein.

The compound according to claim 1 , wherein each of A, D, E, G, J, L, M, and Q are C; each of R 1 and R 2 is independently selected from H or halo; R 4 is H or (C 1 -C 6 )alkyl, R 3 and R 5 are H; R 6 and R 7 are linked to each other as a 4-12 membered mono- or bicyclic urea ring; R 8 , R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , a, b, c, d, e, and f are as defined herein.

The compound according to claim 1 , wherein the compound is the single enantiomer bearing an (R)-configuration at C-3, each of A, D, E, G, J, L, M, and Q are C; each of R 1 and R 2 is independently selected from H or halo; R 4 is H or (C 1 -C 6 )alkyl, R 3 and R 5 are H; R 6 and R 7 are linked to each other as a 4-12 membered mono- or bicyclic amide ring; R 8 , R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , a, b, c, d, e, and f are as defined herein.

The compound according to claim 1 , wherein the compound is the single enantiomer bearing an (R)-configuration at C-3, each of A, D, E, G, J, L, M, and Q are C; each of R 1 and R 2 is independently selected from H or halo; R 4 is H or (C 1 -C 6 )alkyl, R 3 and R 5 are H; R 6 and R 7 are linked to each other as a 4-12 membered mono- or bicyclic urea ring; R 8 , R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , a, b, c, d, e, and f are as defined herein.
6 . A pharmaceutical composition comprising a compound according to claim 1 , or a pharmaceutically acceptable salt or hydrate thereof, and a pharmaceutically acceptable carrier, adjuvant, or diluent.
7 . A method of treating a Cry-mediated disease or disorder in a subject, comprising administering to the subject a therapeutically effective amount of the pharmaceutical composition according to claim 6 .
8 . A method of alleviating a symptom of a Cry-mediated disease or disorder in a subject, comprising administering to the subject a therapeutically effective amount of the pharmaceutical composition according to claim 6 .
9 . A method of monitoring progression or prognosis of a Cry-mediated disease or disorder in a subject, comprising:
measuring an effective amount of one or more cryptochromes or cryptochrome-regulated genes in a first sample from the subject at a first period of time; measuring an effective amount of one or more cryptochromes or cryptochrome-regulated genes in a second sample from the subject at a second period of time; and comparing the amount of the one or more cryptochromes or cryptochrome-regulated genes detected in the first sample to the amount of the one or more cryptochromes or cryptochrome-regulated genes detected in the second sample, or to a reference value.Join the waitlist — get patent alerts
