Abstract

Disclosed are diagnostics kits and compositions for neurodegenerative diseases, and in particular to design ankyrin repeat protein (DARPins) reagents that distinguish Alzheimer's disease (AD) from Parkinson's disease (PD). Methods of diagnosing, monitoring treatment efficacy and developing treatments for neurodegenerative diseases, such as AD and PD are disclosed based upon the use of the AD or PD specific DARPins.

What is claimed is: 
     
         1 . A kit for detecting a neurodegenerative disease or monitoring the efficacy of a neurodegenerative treatment, comprising at least one molecule capable of detecting at least one neurodegenerative-associated design ankyrin repeat protein (DARPin) and directions for using the kit. 
     
     
         2 . The kit of  claim 1 , further comprising at least one neurodegenerative-associated DARPin. 
     
     
         3 . The kit of  claim 2 , wherein the at least one neurodegenerative-associated DARPin is specific for detecting Alzheimer's Disease (AD) and/or Parkinson's Disease (PD). 
     
     
         4 . The kit of  claim 3 , wherein the at least one DARPin specific for detecting AD is selected from at least one member of the group consisting of: ADC1 (SEQ ID NO: 5), ADC3 (SEQ ID NO: 7), ADC6 (SEQ ID NO: 9), ADC7 (SEQ ID NO: 11). 
     
     
         5 . The kit of  claim 3 , wherein the at least one DARPin specific for detecting PD is selected from at least one member of the group consisting of: PDA6 (SEQ ID NO: 13), PDA8 (SEQ ID NO: 15), PDA9 (SEQ ID NO: 17), and PDC1 (SEQ ID NO: 19). 
     
     
         6 . The kit of  claim 2 , further comprising:
 at least one positive control; and   at least one negative control.   
     
     
         7 . A kit for detecting a neurodegenerative disease or monitoring the efficacy of a neurodegenerative treatment, the kit comprising at least one neurodegenerative-associated design ankyrin repeat protein (DARPin). 
     
     
         8 . The kit of  claim 7 , wherein the at least one neurodegenerative-associated DARPin is specific for detecting Alzheimer's Disease (AD) and/or Parkinson's Disease (PD). 
     
     
         9 . The kit of  claim 8 , wherein the at least one DARPin specific for detecting AD is selected from at least one member of the group consisting of: ADC1 (SEQ ID NO: 5), ADC3 (SEQ ID NO: 7), ADC6 (SEQ ID NO: 9), ADC7 (SEQ ID NO: 11). 
     
     
         10 . The kit of  claim 8 , wherein the at least one DARPin specific for detecting PD is selected from at least one member of the group consisting of: PDA6 (SEQ ID NO: 13), PDA8 (SEQ ID NO: 15), PDA9 (SEQ ID NO: 17), and PDC1 (SEQ ID NO: 19). 
     
     
         11 . The kit of  claim 7 , further comprising:
 at least one positive control;   at least one negative control; and   at least one antibody capable of binding to the at least one neurodegenerative-associated DARPin.   
     
     
         12 . The kit of  claim 7 , wherein the kit in an ELISA kit. 
     
     
         13 . The kit of  claim 11 , wherein the at least one antibody capable of binding to the at least one neurodegenerative-associated DARPin is a monoclonal antibody. 
     
     
         14 . The kit of  claim 13 , wherein the monoclonal antibody has an affinity of the DARPin of 0.5 μM. 
     
     
         15 . The kit of  claim 11 , further comprising a sample capture device, wherein the sample capture device contains the at least one neurodegenerative-associated DARPin, the at least one positive control, the at least one negative control, and the at least one antibody capable of binding to the at least one neurodegenerative-associated DARPin. 
     
     
         16 . A composition comprising at least one neurodegenerative-associated design ankyrin repeat protein (DARPin) specific for detecting Alzheimer's Disease (AD) and/or Parkinson's Disease (PD). 
     
     
         17 . The composition of  claim 16 , wherein the at least one DARPin specific for detecting AD is selected from at least one member of the group consisting of: ADC1 (SEQ ID NO: 5), ADC3 (SEQ ID NO: 7), ADC6 (SEQ ID NO: 9), ADC7 (SEQ ID NO: 11). 
     
     
         18 . The composition of  claim 16 , wherein the at least one DARPin specific for detecting PD is selected from at least one member of the group consisting of: PDA6 (SEQ ID NO: 13), PDA8 (SEQ ID NO: 15), PDA9 (SEQ ID NO: 17), and PDC1 (SEQ ID NO: 19). 
     
     
         19 . The composition of  claim 16 , further comprising at least one antibody capable of binding to the at least one neurodegenerative-associated DARPin. 
     
     
         20 . The composition of  claim 19 , further comprising
 at least one positive control; and   at least one negative control.
