Abstract

Highly bioavailable compositions and related methods of improving the bioavailability of one or more compounds are disclosed. The compositions and methods disclosed herein may be employed to improve the bioavailability of poorly soluble or poorly bioavailable ingredients (e.g., curcumin) in a subject (e.g., a mammal).

What is claimed is: 
     
         1 . A composition comprising curcumin and one or more dietary ingredient compounds, one or more surfactant and one or more lipid carriers, wherein the one or more lipid carriers comprise a single chain conjugated lipid carrier, and wherein the composition has at least about 1,250-fold higher bioavailability relative to an unformulated compound. 
     
     
         2 . The composition of  claim 1 , wherein the single chain conjugated lipid carrier further comprise a single chain conjugated lipid carrier selected from the group consisting of lauroyl macrogol-32 glycerides, caprylocaproyl macrogol-8 glycerides and sodium oleate. 
     
     
         3 . The composition of  claim 1 , wherein the single chain conjugated lipid carriers comprise about 10-80% w/w of the composition. 
     
     
         4 . The composition of  claim 1 , wherein the one or more dietary ingredient compounds comprise about 1-30% w/w of the composition. 
     
     
         5 . The composition of  claim 1 , wherein the one or more surfactants comprise about 10-90% w/w of the composition. 
     
     
         6 . The composition of  claim 1 , wherein the one or more surfactants and the single chain conjugated lipid carrier each have an HLB greater than 10. 
     
     
         7 . position of  claim 1 , wherein the one or more surfactants further comprise a surfactant from the group consisting of polysorbate 20, polysorbate 80, Cremophor RH40, Solutol, Cremophor EL, Span 20, Cremophor RH and Brij.

A composition comprising curcumin and one or more dietary ingredient compounds, one or more surfactant and one or more lipid carriers, wherein the one or more lipid comprise a single chain conjugated lipid carrier having a hydrophilic-lipophilic balance (HLB) greater than 10, and wherein the composition has at least about 1,250-fold higher bioavailability relative to an unformulated compound.

The composition of  claim 8 , wherein the one or more lipid carriers further comprise a lipid carrier selected from the group consisting of Gelucire 44/14, Gelucire 33/01, Gelucire 50/13, Capryol 90, Peceol, Na Palmitate, Na Oleate, Acconon MC-8-2, Acconon C 44, Acconon CC-6, Vitamin E TPGS, Labrasol and Tea Oleate. 
     
     
         10 . The composition of  claim 8 , wherein the one or more lipid carriers comprise about 10-80% w/w of the composition. 
     
     
         11 . The composition of  claim 8 , wherein the one or more dietary ingredient compounds comprise about 1-30% w/w of the composition. 
     
     
         12 . The composition of  claim 8 , wherein the one or more surfactants comprise about 10-90% w/w of the composition. 
     
     
         13 . The composition of  claim 8 , wherein the one or more surfactants have an HLB greater than 10. 
     
     
         14 . The composition of  claim 8 , wherein the one or more surfactants further comprise a surfactant from the group consisting of polysorbate 20, polysorbate 80, Cremophor RH40, Solutol, Cremophor EL, Span 20, Cremophor RH and Brij. 
     
     
         15 . The composition of  claim 8 , wherein the single chain conjugated lipid carrier further comprise a single chain conjugated lipid carrier selected from the group consisting of lauroyl macrogol-32 glycerides, caprylocaproyl macrogol-8 glycerides and sodium oleate.

A composition comprising curcumin and one or more dietary ingredient compounds, one or more surfactant and one or more lipid carriers, wherein the one or more lipid carriers comprise a single chain conjugated lipid carrier, wherein the one or more surfactants have an HLB greater than 10, and wherein the composition has at least about 1,250-fold higher bioavailability relative to an unformulated compound.

The composition of  claim 16 , wherein the one or more lipid carriers further comprise a lipid carrier selected from the group consisting of Gelucire 44/14, Gelucire 33/01, Gelucire 50/13, Capryol 90, Peceol, Na Palmitate, Na Oleate, Acconon MC-8-2, Acconon C 44, Acconon CC-6, Vitamin E TPGS, Labrasol and Tea Oleate. 
     
     
         18 . The composition of  claim 16 , wherein the one or more lipid carriers comprise about 10-80% w/w of the composition. 
     
     
         19 . The composition of  claim 16 , wherein the one or more dietary ingredient compounds comprise about 1-30% w/w of the composition. 
     
     
         20 . The composition of  claim 16 , wherein the one or more surfactants comprise about 10-90% w/w of the composition. 
     
     
         21 . The composition of  claim 16 , wherein the one or more surfactants further comprise a surfactant from the group consisting of polysorbate 20, polysorbate 80, Cremophor RH40, Solutol, Cremophor EL, Span 20, Cremophor RH and Brij.
