Abstract
This invention relates to methods for promoting reinnervation of auditory hair cells, specifically, by inhibiting Repulsive Guidance Molecule a (RGMa), a repulsive axonal guidance molecule that is expressed in the cochlea, or its receptor, neogenin.

A method of promoting or enhancing auditory neuron innervation of auditory hair cells, the method comprising administering to the auditory neurons an effective amount of an inhibitor of neogenin, wherein the inhibitor is an inhibitory nucleic acid that specifically reduces expression of neogenin.
12 . The method of claim 11 , wherein the inhibitory nucleic acid is antisense or siRNA.
13 . The method of claim 11 , further comprising administering one or more agents that promote the proliferation, differentiation, or survival of sensory neurons.
14 . The method of claim 13 , wherein the one or more agents are selected from the group consisting of caspase inhibitors, retinoic acid, Bone Morphogenetic Protein 4 (BMP4), anti-oxidants/Nrf2 activators, brain-derived neurotrophic factor (BDNF), neurotrophin 3 (NT3), and NT4/5.

A method of promoting or enhancing innervation of auditory hair cells in the inner ear of a subject in need thereof, the method comprising administering to the inner ear of the subject an effective amount of an inhibitor of neogenin, wherein the inhibitor is an inhibitory nucleic acid that specifically reduces expression of neogenin.
16 . The method of claim 15 , wherein the inhibitory nucleic acid is antisense or siRNA.
17 . The method of claim 15 , further comprising administering one or more agents that promote the proliferation, differentiation, or survival of sensory neurons.
18 . The method of claim 17 , wherein the one or more agents are selected from the group consisting of caspase inhibitors, retinoic acid, BMP4, anti-oxidants/Nrf2 activators, BDNF, NT3, and NT4/5.
19 . The method of claim 15 , wherein the subject has a hearing disorder that results from aging, or has been or will be exposed to an ototoxic level of noise or to one or more ototoxic agents.
20 . The method of claim 19 , wherein the subject has been or will be exposed to one or more ototoxic agents, and wherein the one or more ototoxic agents is radiation, antibiotics, chemotherapeutic agents, anti-inflammatory drugs, heavy metals, or other known or suspected ototoxic agents.
21 . The method of claim 15 , further comprising administering to the inner ear of the subject one or more neural progenitor cells.
22 . A method of treating a subject who has or is likely to have hearing loss or a balance disorder as a result of loss of or decrease in innervation of auditory hair cells, the method comprising administering to the inner ear of the subject an effective amount of an inhibitor of neogenin, wherein the inhibitor is an inhibitory nucleic acid that specifically reduces expression of neogenin.
23 . The method of claim 22 , wherein the inhibitory nucleic acid is antisense or siRNA.
24 . The method of claim 22 , further comprising administering one or more agents that promote the proliferation, differentiation, or survival of sensory neurons.
25 . The method of claim 24 , wherein the one or more agents are selected from the group consisting of caspase inhibitors, retinoic acid, BMP4, anti-oxidants/Nrf2 activators, BDNF, NT3, and NT4/5.
26 . The method of claim 22 , wherein the subject has a hearing disorder that results from aging, or has been or will be exposed to an ototoxic level of noise or to one or more ototoxic agents
27 . The method of claim 26 , wherein the subject has been or will be exposed to one or more ototoxic agents, and wherein the one or more ototoxic agents is radiation, antibiotics, chemotherapeutic agents, anti-inflammatory drugs, heavy metals, or other known or suspected ototoxic agents.
