Abstract
Heteroaryl pyridone and aza-pyridone compounds are provided, including stereoisomers, tautomers, and pharmaceutically acceptable salts thereof, useful for inhibiting Btk kinase, and for treating immune disorders such as inflammation mediated by Btk kinase. Methods of using heteroaryl pyridone and aza-pyridone compounds for in vitro, in situ, and in vivo diagnosis, and treatment of such disorders in mammalian cells, or associated pathological conditions, are disclosed.

2 . A method of treating urticaria, the method comprising administering a pharmaceutical composition to a patient having urticaria, wherein the pharmaceutical composition comprises a therapeutically effective amount of a compound selected from the group consisting of:

The method of claim 2 , further comprising administering an additional therapeutic agent or agents selected from an anti-inflammatory agent, an immunomodulatory agent, chemotherapeutic agent, an apoptosis-enhancer, a neurotropic factor, an agent for treating cardiovascular disease, an agent for treating liver disease, an anti-viral agent, an agent for treating blood disorders, an agent for treating diabetes, and an agent for treating immunodeficiency disorders.
4 . The method of claim 3 , wherein the additional therapeutic agent is an anti-inflammatory agent.
5 . The method of claim 4 , wherein the anti-inflammatory agent is a non-steroidal anti-inflammatory drug.
6 . The method of claim 3 , wherein the additional therapeutic agent is an immunomodulatory agent.
7 . The method of claim 2 , wherein the compound is 2-{3′-hydroxymethyl-1-methyl-5-[5-((S)-2-methyl-4-oxetan-3-yl-piperazin-1-yl)-pyridin-2-ylamino]-6-oxo-1,6-dihydro-[3,4′]bipyridinyl-2′-yl}-7,7-dimethyl-3,4,7,8-tetrahydro-2H,6H-cyclopenta[4,5]pyrrolo[1,2-a]pyrazin-1-one, or a pharmaceutically acceptable salt thereof.
8 . The method of claim 7 , wherein the patient is a human.
9 . The method of claim 8 , wherein the pharmaceutical composition is administered orally in the form of a capsule or tablet.
10 . The method of claim 9 , wherein the pharmaceutical composition is administered in an amount of about 10 mg to about 1000 mg daily.
11 . The method of claim 9 , wherein the pharmaceutical composition is administered in an amount of about 100 mg to about 300 mg daily.
12 . The method of claim 10 , wherein the pharmaceutical composition is administered once daily.
13 . The method of claim 10 , wherein the pharmaceutical composition is administered twice daily.
14 . The method of claim 11 , wherein the pharmaceutical composition is administered once daily.
