Claims

Abstract

Provided herein are trispecific antibodies that include a) a first monomer, b) a second monomer, and c) a light chain. The first monomer includes a first variant Fc domain and a variable heavy domain, the second monomer includes a second variant Fc domain and a first scFv domain and the light chain includes a variable light domain. The variable heavy domain and the variable light domain form an antigen binding domain. The trispecific antibody further includes a second scFv domain on either the first or second monomers.

What is claimed is: 
     
         1 . A trispecific antibody comprising:
 a) a first monomer comprising a first heavy chain comprising a first variant Fc domain and a first variable heavy domain;   b) a second monomer comprising:
 i) a second variant Fc domain; 
 ii) a first scFv domain; and 
   c) a light chain comprising a constant light domain and a variable light domain;   wherein the first variable heavy domain and the first variable light domain form an antigen binding domain, and   wherein one of the first and second monomers further comprises a second scFv domain.   
     
     
         2 . The trispecific antibody of  claim 1 , wherein the first monomer comprises the second scFv domain. 
     
     
         3 . The trispecific antibody of  claim 1 , wherein the second monomer comprises the second scFv domain. 
     
     
         4 . A nucleic acid composition comprising:
 a) a first nucleic acid encoding the first monomer of  claim 1 ;   b) a second nucleic acid encoding the second monomer of  claim 1 ; and   c) a third nucleic acid encoding the third monomer of  claim 1 .   
     
     
         5 . An expression vector composition comprising:
 a) a first expression vector comprising the first nucleic acid of  claim 4 ;   b) a second expression vector comprising the second nucleic acid of  claim 4 ; and   c) a third expression vector comprising the third nucleic acid of  claim 4 .   
     
     
         6 . A host cell comprising the expression vector composition of  claim 5 . 
     
     
         7 . A method of making a trispecific antibody comprising culturing the host cell of  claim 6  under conditions wherein the trispecific antibody is expressed and recovering the trispecific antibody.
