Abstract
Provided herein are methods of applying the ERH gene in the preparation of bladder cancer diagnosis and treatment products. The ERH gene is found to have a good correlation with bladder cancer. In addition, an RNA interference vector is constructed to perform cell function experiments to study the effect of ERH gene on the proliferation and clonality of bladder cancer cells. The methods disclosed herein provide a research basis for clinical diagnosis and treatment of bladder cancer and has a good application prospect.

7 . A method for screening a compound for treating or preventing bladder cancer, comprising:
(1) contacting a cell that expresses a sequence that is at least 80% homologous to an ERH nucleotide sequence with a plurality of compounds; (2) selecting a compound that causes the cell to exhibit a reduced level of expression of a sequence having at least 80% homology with the ERH nucleotide sequence expressed by the cell.

12 . A method of using an ERH gene inhibitor or ERH inhibitor in preparation of anti-bladder cancer compounds, comprising:
(1) providing a polypeptide that comprises at least 80% homology to an ERH nucleotide sequence; (2) detecting binding activity between a compound to be tested and the polypeptide; and (3) selecting a compound that binds to the polypeptide.

The method of claim 12 , wherein the anti-bladder cancer compounds exhibit one or more of the following activities to inhibit the expression level of ERH in bladder cancer cells: activating an inhibitor gene of the ERH gene, activating a protein that inhibits expression of an ERH gene, introducing an siRNA that inhibits ERH expression, introducing an antisense oligonucleotide that inhibits ERH expression, introducing a vector that inhibits ERH expression, and activating a microRNA that promotes degradation of ERH mRNA, introducing molecules that promote degradation of ERH proteins, suppressing the expression of factors and proteins that promote the expression level of ERH genes, inhibiting the activity of promoter or enhancer of ERH genes, enhancing the activity of ERH gene silencer, and introducing antibodies or immunologically active fragments that bind to ERH proteins.
14 . The method of claim 12 , wherein the ERH gene inhibitor or ERH protein inhibitor is interfering RNA, and the target sequence of the interfering RNA is SEQ ID NO: 15.

16 . The method of claim 12 , wherein the inhibitors inhibit proliferation or metastasis of bladder cancer cells or the formation of tumors.

A composition for treating or preventing bladder cancer, comprising a pharmaceutically effective amount of an antisense oligonucleotide or interfering RNA targeting ERH as an active ingredient or a pharmaceutically effective amount of an antibody or an immunologically active fragment that binds to an ERH protein, and further includes pharmaceutically acceptable carriers.
18 . A diagnostic reagent for bladder cancer, comprising primers for specifically amplifying the ERH gene or probes hybridized with the nucleic acid sequence of the ERH gene; or antibodies or immunologically active fragments that bind to ERH proteins.
