Abstract

The present invention relates to immunoglobulins that bind FcγRIIb+ cells and coengage the antigen on the cell's surface and an FcγRIIb on the cell's surface, methods for their generation, and methods for using the immunoglobulins.

         21 . A method of treating cancer in a patient in need of such treatment, comprising:
 administering to the patient an anti-human CD40 antibody comprising a light chain and a heavy chain, the heavy chain comprising a human IgG1 Fc domain comprising a S267E amino acid substitution, wherein the numbering is according to the EU index as in Kabat.   
     
     
         22 . The method of  claim 21 , wherein the cancer is pancreatic cancer. 
     
     
         23 . The method of  claim 21 , comprising administering the anti-human CD40 antibody in combination with one or more chemotherapeutic agents. 
     
     
         24 . The method of  claim 23 , wherein the chemotherapeutic agent is gemcitabine. 
     
     
         25 . The method of  claim 24 , wherein the one or more chemotherapeutic agents are gemcitabine in combination with paclitaxel. 
     
     
         26 . The method of  claim 24 , comprising administering the anti-human CD40 antibody in combination with a second antibody. 
     
     
         27 . The method of  claim 25 , comprising administering the anti-human CD40 antibody in combination with a second antibody. 
     
     
         28 . A method of causing B cell proliferation in a patient comprising:
 contacting the B cell of the patient with an anti-human CD40 antibody comprising amino acid substitution S267E in the Fc region as compared to a parent human IgG1 Fc region, wherein the numbering is according to the EU index as in Kabat thereby causing B cell proliferation in the patient.
