Amino quinazolines as kinase inhibitors

Abstract

Disclosed are compounds having the formula: wherein R 1 , R 2 , R 3 , and Z are as defined herein, and methods of making and using the same.

What is claimed is: 
     
         1 . A compound which is 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol having the structure of 
       
         
           
           
               
               
           
         
         or a pharmaceutically acceptable salt thereof. 
       
     
     
         2 . The compound according to  claim 1 , which is a pharmaceutically acceptable salt of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol. 
     
     
         3 . The compound according to  claim 1 , which is 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol. 
     
     
         4 . A pharmaceutical composition comprising the compound of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol having the structure of 
       
         
           
           
               
               
           
         
         or a pharmaceutically acceptable salt thereof, 
         and a pharmaceutically acceptable excipient. 
       
     
     
         5 . The pharmaceutical composition according to  claim 4 , comprising the compound of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol. 
     
     
         6 . The pharmaceutical composition according to  claim 4 , comprising a pharmaceutically acceptable salt of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol. 
     
     
         7 . A method of treating a disease mediated by RIP2 kinase comprising administering to a human in need thereof a therapeutically effective amount of the compound of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol having the structure of 
       
         
           
           
               
               
           
         
         or a pharmaceutically acceptable salt thereof, 
         wherein the disease is selected from the group consisting of uveitis, dermatitis, acute lung injury, type 2 diabetes mellitus, arthritis, rheumatoid arthritis, ulcerative colitis, Crohn's disease, early-onset inflammatory bowel disease, extraintestinal inflammatory bowel disease, prevention of ischemia reperfusion injury in solid organ transplant, non-alcohol steatohepatitis, autoimmune hepatitis, asthma, systemic lupus erythematosus, multiple sclerosis, sarcoidosis, Blau syndrome/early-onset sarcoidosis, Wegner's granulomatosis, and interstitial pulmonary disease. 
       
     
     
         8 . The method according to  claim 7  comprising administering to a human in need thereof, a therapeutically effective amount of the compound of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol. 
     
     
         9 . The method according to  claim 7 , wherein the disease is selected from the group consisting of uveitis, Blau Syndrome, early-onset sarcoidosis, ulcerative colitis, Crohn's disease, Wegener's granulamatosis and sarcoidosis. 
     
     
         10 . The method according to  claim 7 , wherein the disease is Crohn's disease. 
     
     
         11 . The method according to  claim 7 , wherein the disease is ulcerative colitis. 
     
     
         12 . The method according to  claim 7 , wherein the disease is Blau syndrome. 
     
     
         13 . The method according to  claim 7 , wherein the disease is rheumatoid arthritis. 
     
     
         14 . A method of treating a disease selected from the group consisting of Blau Syndrome, ulcerative colitis, Crohn's disease, and rheumatoid arthritis comprising administering the compound of 2-((4-(benzo[d]thiazol-5-ylamino)-6-(tert-butylsulfonyl)quinazolin-7-yl)oxy)ethanol to a human in need thereof.
