Abstract
This disclosure provides generally for antimicrobial compositions and methods of use comprising an anthocyanin, an anthocyanidin or metabolites thereof. Methods for promoting healing of a wound using these compositions are also disclosed. These compositions have broad spectrum antimicrobial activity and are safe for human and animal uses. Further, these compositions are safe for medical uses and industrial uses as antiseptic preparations to reduce or prevent microbial growth.

1 . A therapeutic wound healing composition, comprising a wound healing agent selected from the group consisting of: protocatechuic acid or a combination of protocatechuic acid and 2,4,6 trihydroxybenzaldehyde, wherein the wound healing agent is present in an amount sufficient to promote healing of a wound.
2 . The composition of claim 1 , wherein the wound healing agent is present in an amount to promote a wound healing response selected from the group consisting of optimizing and modulating a production of local growth hormones, controlling inflammation, and killing bacteria.
3 . The composition of claim 1 , wherein the wound healing agent is present in an amount to elicit one or more wound healing responses selected from:
a. reducing at least one microbe selected from the group consisting of L. casei, C. difficile, P. acnes, C. perfringens, L. casei, E. coli, S. aureus 6538, S. aureus 33591, S. mutans, S. pyogenes, P. aeruginosa, K. pneumonia, and C. albicans; b. increasing localized IGF-1 expression; c. reducing localized TGF-E expression; and d. reducing localized EGF expression.
4 . The composition of claim 1 , wherein the wound is a burn, skin break, bone break, muscle tear, puncture, surgical incision site, microdermabrasion site, skin graft site, a wound associated with diabetes, a bedsore, a pressure sore, or a laceration.
5 . The composition according to claim 1 , wherein the wound healing agent is present at a concentration of at least 25 mM.
6 . The composition of claim 1 , wherein the wound healing agent is present at a concentration of between 20 to 500 mM.
7 . The composition of claim 1 , wherein the wound healing agent is present at a concentration of between 10 to 200 mM.
8 . The composition of claim 1 , wherein the wound healing agent is a combination of protocatechuic acid and 2,4,6 trihydroxybenzaldehyde.
9 . The composition of claim 8 , wherein the protocatechuic acid and 2,4,6 trihydroxybenzaldehyde are each present at a concentration of between 10 mM to 200 mM.
10 . The composition of claim 8 , wherein the protocatechuic acid and 2,4,6 trihydroxybenzaldehyde are each present at a concentration of about 50 mM.
11 . The composition of claim 1 , wherein the composition is a liquid, solution, cream, ointment, salve, gel, emulsion, suspension, dispersion, or paste.
12 . The composition of claim 1 , wherein the composition is in a form of ultradeformable liposomes.
13 . The composition of claim 1 , wherein the wound healing agent is dissolved in a solution.
14 . The composition of claim 13 , wherein the solution comprises water, isopropyl alcohol, propylene glycol, an oil, or a mixture thereof.
15 . The composition of claim 1 , wherein the composition is formulated for oral, topical, buccal, injection, inhalation, sublingual, or transdermal administration.
16 . The composition of claim 1 , wherein the composition is formulated for topical, oral or transdermal administration or intradermal or subcutaneous injection to a skin tissue; wherein the composition promotes an effect selected from the group consisting of reducing inflammation, optimizing growth hormone production, and increasing collagen stimulation.
17 . A wound healing composition having an enhanced ability to prevent microbial infection and improve healing comprising (a) the composition according to claim 1 and (b) a medicament useful for treating wounds.
18 . An antiseptic composition comprising: an effective amount of an anthocyanin or anthocyanidin or anthocyanin metabolite or anthocyanidin metabolite consisting of a cyanidin-3-glucoside, protocatechuic acid, or 2,4,6 trihydroxybenzaldehyde or a mixture thereof.
19 . The antiseptic composition of claim 18 , wherein the anthocyanin or anthocyanidin or anthocyanin or anthocyanidin metabolites is present at a concentration of between 20 to 500 mM.
