Transdermal systems containing multilayer adhesive matrices to modify drug delivery

Abstract

The first polymeric adhesive matrix can release the drug more quickly or more slowly than the second polymeric adhesive matrix. Through the selection of the two matrices, the delivery profile of the drug through the skin can be selectively modified and controlled.

         48 . A transdermal drug-containing dosage unit comprising:
 a. a backing layer substantially impervious to the drug to be delivered transdermally;   b. a polymeric adhesive system comprising:
 i. a drug consisting of estradiol in an amount from 1.25% to about 4.0% w/w, 
 ii. an acrylic adhesive, 
 iii. a silicon adhesive, 
 iv. povidone, and 
 v. a solvent, wherein the polymeric adhesive system has a thickness of at least about 100 gsm; and 
   c. a release liner in contact with the polymeric adhesive system.   
     
     
         49 . The transdermal drug-containing dosage unit of  claim 48 , wherein the estradiol is estradiol hemihydrate. 
     
     
         50 . The transdermal drug-containing dosage unit of  claim 48 , wherein the polymeric adhesive system has a thickness of about 125 gsm. 
     
     
         51 . The transdermal drug-containing dosage unit of  claim 48 , wherein the polymeric adhesive system has a thickness of about 150 gsm. 
     
     
         52 . The transdermal drug-containing dosage unit of  claim 48 , wherein a thickness of the backing layer and a thickness of the polymeric adhesive system can be modified to change a drug delivery rate of the drug-containing dosage unit. 
     
     
         53 . A transdermal drug-containing dosage unit comprising:
 a. a backing layer substantially impervious to the drug to be delivered transdermally;   b. a polymeric adhesive system comprising:
 i. a drug consisting of estradiol in an amount from about 0.1% to about 4.0% w/w, 
 ii. an acrylic adhesive, 
 iii. a silicon adhesive, 
 iv. povidone, and 
 v. a solvent, wherein the polymeric adhesive system has a thickness of between about 5 and 150 gsm; and 
   c. a release liner in contact with the polymeric adhesive system.
