Abstract

Disclosed are nutritional compositions including human milk oligosaccharides in combination with long chain polyunsaturated fatty acids and/or carotenoids that can be administered to preterm infants, term infants, toddlers, and children for reducing inflammation and the incidence of inflammatory diseases.

A nutritional powder comprising a human milk oligosaccharide selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, 3-fucosyllactose, lacto-N-tetraose, lacto-N-neotetraose, lacto-N-fucopentaose, and combinations thereof and docosahexaenoic acid in an amount of about 3 mg per 100 kcals to about 16 mg per 100 kcals. 
     
     
         2 . The nutritional powder of  claim 1 , wherein the nutritional powder further comprises arachidonic acid in an amount of about 10 mg per 100 kcals to about 31 mg per 100 kcals. 
     
     
         3 . The nutritional powder of  claim 1 , wherein the human milk oligosaccharide is present in an amount of from about 0.01 mg/mL to about 20 mg/mL and is selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, lacto-N-neotetraose, and combinations thereof. 
     
     
         4 . The nutritional powder of  claim 1 , wherein the nutritional powder further comprises a long chain polyunsaturated fatty acid selected from the group consisting of linoleic acid, linolenic acid, arachidonic acid, eicosapentaenoic acid, docosapentaenoic acid, and combinations thereof. 
     
     
         5 . The nutritional powder of  claim 1 , wherein the human milk oligosaccharide is derived from a fermentation process, an enzymatic process, a chemical process, or a combination thereof. 
     
     
         6 . The nutritional powder of  claim 1 , wherein the human milk oligosaccharide is 6′-sialyllactose. 
     
     
         7 . The nutritional powder of  claim 1 , wherein the human milk oligosaccharide is 2′-fucosyllactose. 
     
     
         8 . The nutritional powder of  claim 7 , wherein the 2′-fucosyllactose is present in a concentration of about 0.01 mg/mL to less than 2 mg/mL. 
     
     
         9 . The nutritional powder of  claim 1 , further comprising about 0.001 μg/mL to about 5 μg/mL of a carotenoid.

         11 . The nutritional powder of  claim 9 , wherein the carotenoid is selected from the group consisting of lutein, lycopene, beta-carotene, and combinations thereof. 
     
     
         12 . The nutritional powder of  claim 1  further comprising a nucleotide in an amount of from about 10 mg/L to about 200 mg/L. 
     
     
         13 . The nutritional powder of  claim 12 , wherein the nucleotide is present in an amount of from about 42 mg/L to about 102 mg/L.

A nutritional powder comprising a human milk oligosaccharide selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, 3-fucosyllactose, lacto-N-tetraose, lacto-N-neotetraose, lacto-N-fucopentaose, and combinations thereof, docosahexaenoic acid in an amount of about 3 mg per 100 kcals to about 16 mg per 100 kcals, and a carotenoid in an amount of about 0.001 μg/mL to about 5 μg/mL. 
     
     
         15 . The nutritional powder of  claim 14 , wherein the human milk oligosaccharide is selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, lacto-N-neotetraose, and combinations thereof. 
     
     
         16 . The nutritional powder of  claim 14 , wherein the human milk oligosaccharide is derived from a source selected from human, bovine, ovine, porcine, caprine, or combinations thereof. 
     
     
         17 . The nutritional powder of  claim 14  further comprising a nucleotide in an amount of about 10 mg/L to about 200 mg/L. 
     
     
         18 . The nutritional powder of  claim 14 , wherein the nutritional powder further comprises arachidonic acid in an amount of about 10 mg per 100 kcals to about 31 mg per 100 kcals.

A method of modulating immunity in an infant, toddler, or child in need thereof, the method comprising administering to the infant, child or toddler a nutritional powder comprising a human milk oligosaccharide, docosahexaenoic acid in an amount of from about 3 mg per 100 kcals to about 16 mg per 100 kcals, and from about 0.001 μg/mL to about 5 μg/mL of a carotenoid. 
     
     
         23 . The method of  claim 22 , wherein the nutritional powder further comprises arachidonic acid in an amount of about 10 mg per 100 kcals to about 31 mg per 100 kcals. 
     
     
         24 . The method of  claim 22 , wherein the human milk oligosaccharide is selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, 3-fucosyllactose, lacto-N-tetraose, lacto-N-neotetraose, lacto-N-fucopentaose, and combinations thereof. 
     
     
         25 . The method of  claim 22 , wherein the human milk oligosaccharide is selected from the group consisting of 6′-sialyllactose, 3′-sialyllactose, 2′-fucosyllactose, lacto-N-neotetraose, and combinations thereof.

         30 . The nutritional powder of  claim 29  further comprising a nucleotide in an amount of about 10 mg/L to about 200 mg/L.
