Fc VARIANTS WITH ALTERED BINDING TO FcRn

Abstract

The present application relates to optimized IgG immunoglobulin variants, engineering methods for their generation, and their application, particularly for therapeutic purposes.

         25 . A method of producing a polypeptide comprising a variant Fc region as compared to a parent Fc region, said variant Fc region comprising amino acid substitutions selected from the group consisting of: N434S, V308F, V308C, V308W, V308Y, M428L/N434S, V259I/V308F, and V259I/V308F/M428L, wherein numbering is according to the EU Index in Kabat et al., said method comprising providing a cell comprising a nucleic acid encoding said polypeptide, wherein said cell is cultured under conditions suitable for expression of said polypeptide. 
     
     
         26 . A method according to  claim 25 , wherein said nucleic acid is contained in an expression vector. 
     
     
         27 . A method according to  claim 25 , wherein said variant Fc region comprises M428L/N434S.

A host cell comprising a nucleic acid encoding a polypeptide comprising a variant Fc region as compared to a parent Fc region, said variant Fc region comprising amino acid substitutions selected from the group consisting of: N434S, V308F, V308C, V308W, V308Y, M428L/N434S, V259I/V308F, and V259I/V308F/M428L.
