Analyte Detection
Abstract
The present disclosure provides methods and/or kits for detecting an analyte in a sample. Some embodiments provide a method for detecting a non-nucleic acid analyte in a sample using a solid substrate comprising a bound immobilisation agent and an antibody capture agent and a detectable agent, which can bind to the analyte. The antibody capture agent comprises, at a plurality of sites, a ligand for the immobilisation agent. A complex between the analyte, the antibody capture agent and a detectable agent is formed and immobilised on the solid substrate by binding between the immobilisation agent and the ligand. In some embodiments, the ligand and the immobilisation agent are a binding pair comprising a peptide tag and an anti-peptide tag antibody.

26 . A method for preparing a series of assay kits, comprising:
i) preparing a series of substrates comprising a quantity of antibodies, the antibodies configured to selectively bind with the a certain type of peptide; ii) further preparing a series of solutions comprising a quantity of antibody capture agents covalently conjugated to at least one of the certain type of peptide; and iii) assembling the series of assay kits, the series of assay kits comprising:
a) at least one substrate from the series of substrates, and
b) at least one solution from the series of solutions.
27 . The method of claim 26 , wherein the series of assay kits comprise:
a) a first assay kit comprising a first substrate of the series of substrates and a first solution of the series of solutions, the first solution comprising a quantity of first antibody capture agents; and b) a second assay kit comprising a second substrate of the series of substrates and second solution of the series of solutions, the second solution comprising a quantity of second antibody capture agents.
28 . The method of claim 27 , wherein the quantity of first antibody capture agents comprises a first type of antibody and the quantity of second antibody capture agents comprises a second type of antibody, the first type of antibody different from the second type of antibody.
29 . The method of claim 27 , wherein each first antibody capture agent present in at least a portion of the quantity of first antibody capture agents is covalently conjugated to a plurality of the certain type of peptide.
30 . The method of claim 26 , wherein the quantity of antibodies is obtained from a single batch of antibodies.
31 . The method of claim 26 , wherein the certain type of peptide comprises the amino acid sequence DYKDDDDK (SEQ ID NO: 1).
32 . The method of claim 26 , wherein at least one of the series of substrates is an assay plate.
33 . The method of claim 29 , wherein the assay plate is a multi-well plate.
34 . The method of claim 26 , wherein at least one of the series of substrates is a microfluidic device.
35 . The method of claim 26 , wherein at least one of the series of substrates is a multi-well strip.
36 . The method of claim 26 , wherein at least one of the series of substrates is a bead.
37 . The method of claim 26 , wherein at least one of the series of substrates is a dip stick.
38 . A series of analyte detection kits, comprising:
i) a series of substrates comprising a quantity of anti-peptide antibodies, the anti-peptide antibodies configured to selectively bind with the a certain type of peptide; and ii) a series of solutions comprising quantities of at least two different types of antibody capture agents, each of the at least two different types of antibody capture agents covalently conjugated a plurality of the certain type of peptide.
39 . The series of analyte detection kits of claim 38 , wherein at least one of the series of substrates is a multi-well plate comprising a plurality of reaction vessels coated therein with at least a portion of the quantity of anti-peptide antibodies.
40 . The series of analyte detection kits of claim 38 , wherein at least a portion of the quantity of anti-peptide antibodies is coated at a concentration of 25 μg/ml or less.
41 . The series of analyte detection kits of claim 38 , wherein the series of analyte detection kits are configured to detect analytes in a time of 2 hours or less.
42 . The series of analyte detection kits of claim 38 , wherein the series of analyte detection kits is configured to detect analytes after no more than one washing.
43 . The series of analyte detection kits of claim 38 , comprising a further series of solutions, the further series of solutions comprising detectable agents.
44 . The plural analyte detection kits of claim 43 , wherein the detectable agents comprise detectable tags.
45 . A method for preparing a series of assay kits, comprising:
i) procuring a series of substrates comprising a quantity of antibodies, the antibodies configured to selectively bind with the a certain type of peptide; ii) further procuring a series of solutions comprising a quantity of antibody capture agents covalently conjugated to at least one of the certain type of peptide; and iii) assembling the series of assay kits, each one of the series of assay kits comprising:
a) at least one substrate from the series of substrates, and
