Abstract

Provided herein are methods, compositions, and uses relating to inhibitors of stem cell factor. For example, provided herein are antibodies targeting stem cell factor and methods for treating fibrotic and tissue remodeling diseases.

Claims

exact text as granted — not AI-modified
- 18 . (canceled)

. A biological host capable of producing an antibody or antigen-binding antibody fragment specific for stem cell factor (SCF) isoform b.

. The biological host of claim 19 wherein said biological host is a phage, a bacterium, or a eukaryotic cell.

. The biological host of claim 19 wherein said biological host is a splenocyte or a lymphocyte.

. The biological host of claim 19 wherein said biological host is an E. coli cell, a COS cell, a CHO cell, or a myeloma cell.

. The biological host of claim 19 wherein said biological host comprises a polynucleotide encoding the antibody specific for SCF isoform b.

. The biological host of claim 19 wherein the biological host is a continuous cell line.

. The biological host of claim 19 wherein the biological host is a hybridoma.

. The biological host of claim 19 wherein the biological host is a trioma, B-cell hybridoma, or EBV-hybridoma.

. The biological host of claim 19 wherein the biological host is a mammal

. The biological host of claim 19 wherein the biological host is a mouse, rabbit, goat, sheep, rat, donkey, or hamster.

. The biological host of claim 19 wherein the biological host carries a human immune system.

. The biological host of claim 19 wherein the biological host is a cell isolated from an animal immunized with a peptide comprising an amino acid sequence provided by SEQ ID NO: 4.

. A method of preparing an antibody specific for stem cell factor (SCF) isoform b, the method comprising contacting a host cell with a peptide comprising an amino acid sequence provided by SEQ ID NO: 4.

. The method of claim 31 further comprising isolating an antibody-producing cell that produces the antibody specific for SCF isoform b.

. The method of claim 32 further comprising isolating the antibody specific for SCF isoform b from the antibody-producing cell.

. The method of claim 32 further comprising fusing the antibody-producing cell with a second cell to produce a continuous cell line.

. The method of claim 34 wherein the continuous cell line is a hybridoma.

. The method of claim 31 wherein the peptide is conjugated to an immunogenic carrier or is provided with an adjuvant.

. The method of claim 31 wherein contacting said host cell with said peptide is performed by a method comprising immunizing a host with the peptide.

. The method of claim 37 wherein the host is a mammal

. The method of claim 37 wherein the host is a mouse, rabbit, goat, sheep, rat, donkey, or hamster.

. The method of claim 37 wherein the host carries a human immune system.

. The method of claim 31 wherein the peptide comprises an amino acid from positions 174-202 of SEQ ID NO: 4.

. The method of claim 31 wherein the peptide comprises an amino acid sequence from SEQ ID NO: 1.

. The method of claim 31 wherein the peptide comprises one or more substitutions, deletions, or insertions relative to SEQ ID NO: 4.

. The method of claim 31 further comprising preparing an antigen-binding antibody fragment specific for SCF isoform b from the antibody specific for SCF isoform b.

. The method of claim 31 further comprising isolating a polynucleotide encoding an amino acid sequence of the antibody specific for SCF isoform b.
