Claims

Abstract

The present invention relates to nucleic acid sequences and amino acid sequences which influence bone deposition, the Wnt pathway, ocular development, tooth development, and may bind to LRP. The nucleic acid sequence and polypeptides include Wise and Sost as well as a family of molecules which express a cysteine knot polypeptide. Additionally, the present invention relates to various molecular tools derived from the nucleic acids and polypeptides including vectors, transfected host cells, monochronal antibodies, Fab fragments, and methods for impacting the pathways.

What is claimed is: 
     
         1 . A method for modulating bone deposition comprising contacting a host cell with an antibody to Sost, wherein the antibody prevents wild-type Sost from binding with a Sost binding partner, wherein the Sost binding partner is an LRP having an amino acid sequence selected from the group consisting of SEQ ID NOS: 68, 73, 76, 79, 84, and 86. 
     
     
         2 . The method according to  claim 1 , wherein the antibody is a monoclonal antibody. 
     
     
         3 . The method according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence selected from the group consisting of SEQ ID NOS: 68, 72, and 84. 
     
     
         4 . The method according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 68. 
     
     
         5 . The method according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 72. 
     
     
         6 . The method according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 84. 
     
     
         7 . A composition for modulating bone deposition comprising an antibody to Sost, wherein the antibody prevents wild-type Sost from binding with a Sost binding partner, wherein the Sost binding partner is an LRP having an amino acid sequence selected from the group consisting of SEQ ID NOS: 68, 73, 76, 79, 84, and 86. 
     
     
         8 . The composition according to  claim 1 , wherein the antibody is a monoclonal antibody. 
     
     
         9 . The composition according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence selected from the group consisting of SEQ ID NOS: 68, 72, and 84. 
     
     
         10 . The composition according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 68. 
     
     
         11 . The composition according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 72. 
     
     
         12 . The composition according to  claim 1 , wherein the Sost binding partner is an LRP having an amino acid sequence according to SEQ ID NO: 84.
