Abstract
Provided are monoclonal antibodies that detect CD19 CAR-modified immune cells and CAR-modified immune cells irrespective of the tumor associated antigen they target. Methods of using these functional monoclonal antibodies include, but are not limited to, detection, quantification, activation, and selective propagation of CAR-modified immune cells.

An isolated antibody or antibody fragment thereof that specifically binds to a chimeric antigen receptor (CAR) polypeptide, wherein the isolated antibody or antibody fragment thereof comprises a set of complementarity determining regions (CDRs) HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3, wherein
(a) HCDR1 has the amino acid sequence of SEQ ID NO: 10; (b) HCDR2 has the amino acid sequence of SEQ ID NO: 11; (c) HCDR3 has the amino acid sequence of SEQ ID NO: 12; (d) LCDR1 has the amino acid sequence of SEQ ID NO: 14; (e) LCDR2 has the amino acid sequence of SEQ ID NO: 15; and (f) LCDR3 has the amino acid sequence of SEQ ID NO: 16.
111 . The isolated antibody or antibody fragment thereof of claim 110 , wherein the antibody or antibody fragment thereof is conjugated to a toxin.
112 . A pharmaceutical composition comprising the antibody or antibody fragment thereof of claim 110 .

An isolated antibody or antibody fragment thereof that specifically binds to a chimeric antigen receptor (CAR) polypeptide, wherein the isolated antibody or antibody fragment thereof comprises a heavy chain variable region (VH) having the amino acid sequence of SEQ ID NO: 9 and a light chain variable region (VL) having the amino acid sequence of SEQ ID NO: 13.
114 . The isolated antibody or antibody fragment thereof of claim 113 , wherein the antibody or antibody fragment thereof is conjugated to a toxin.
115 . A pharmaceutical composition comprising the antibody or antibody fragment thereof of claim 113 .

An isolated antibody or antigen-binding fragment thereof that specifically binds to a chimeric antigen receptor (CAR) polypeptide, wherein the antibody or antigen-binding fragment thereof comprises a VH domain comprising an amino acid sequence which is at least 95% identical to the amino acid sequence shown in SEQ ID NO: 9 or a VL domain comprising an amino acid sequence which is at least 95% identical to the amino acid sequence shown in SEQ ID NO: 13.
117 . The antibody of claim 116 , wherein the VL domain comprises three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO: 14, 15, and 16.
118 . The antibody of claim 116 , wherein the VH domain comprises three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO: 10, 11, and 12.
119 . The isolated antibody or antibody fragment thereof of claim 116 , wherein the antibody or antibody fragment thereof is conjugated to a toxin.
