Claims

Abstract

In certain embodiments, this disclosure describes compositions comprising platelet lysates depleted of fibrinogen. In a further embodiment, the composition further comprises a cell culture medium component. This disclosure also provides a method for preparing the composition, comprising the steps of (a) lysing platelets providing a lysate; (b) removing cell debris; and (c) depleting fibrinogen by forming a removable mass by adding a metal salt such as calcium chloride. Furthermore, the disclosure also describes the product produced using said method.

         16 . A method of making fibrinogen depleted platelet lysate comprising
 (a) lysing platelets providing a platelet lysate;   (b) removing cell debris; and   (d) depleting fibrinogen from the platelet lysate under conditions wherein the concentration of fibrinogen is less than 50 μg/ml.   
     
     
         17 . The method of  claim 16 , wherein the cell debris is removed prior to, after or both prior to and after depletion of fibrinogen. 
     
     
         18 . The method of  claim 16 , further comprising pooling multiple platelet lysates prior to depletion of fibrinogen. 
     
     
         19 . The method of  claim 16 , wherein the cell debris is removed by filtration. 
     
     
         20 . The method of  claim 16 , wherein the platelets are lysed by freezing and thawing. 
     
     
         21 . The method of  claim 20 , wherein the freezing and thawing is performed twice. 
     
     
         22 . The method of  claim 16 , wherein the platelets are lysed by mechanical lysis or a lysis buffer. 
     
     
         23 . The method of  claim 16 , further comprising adding a metal salt to the platelet lysate. 
     
     
         24 . The method of  claim 23 , wherein the metal salt is a calcium salt. 
     
     
         25 . The method of  claim 24 , wherein the calcium salt is calcium chloride. 
     
     
         26 . The method of  claim 23 , further comprising adding heparin to the platelet lysate. 
     
     
         27 . The method of  claim 26 , wherein addition of heparin and/or the metal salt results in clot formation. 
     
     
         28 . The method of  claim 27 , further comprising removal of the clot. 
     
     
         29 . The method of  claim 28 , wherein clot removal is performed by centrifugation. 
     
     
         30 . The method of  claim 16 , further comprising lyophilizing the platelet lysate. 
     
     
         31 . The method of  claim 16 , wherein the platelet lysate is added to cell culture media.
