Abstract
The invention relates to a method for sequencing a heteropolymeric target nucleic acid sequence that involves stochastic sensing. The invention also relates to a method for improving a pore for sequencing a target nucleic acid sequence by modifying one or more sites in the pore.

A method for sequencing a heteropolymeric target nucleic acid sequence, comprising: (a) passing the target sequence through a transmembrane pore so that a proportion of the nucleotides in the target sequence interacts one at a time with at least one site in the pore that is capable of discriminating between different nucleotides; and (b) measuring the current passing through the pore during each interaction and thereby determining the sequence of the target sequence.
