Abstract

The invention in suitable embodiments is directed to using a purified, clathrin light chain protein for purposes of drug development, enablement, activity, and/or delivery. In one aspect, a man-made, configurable clathrin light chain composition, formed in whole or in part from isolated, synthetic and or recombinant amino acid residues comprising in whole or in part one or more of a protein sequence, forms one or more type of multifunction nanoscale bio-nanoparticle, such as a biomedical platform, bio-molecular platform, and the like, and using such bio-nanoparticle platforms for development, enablement, and delivery of drugs.

An isolated nanoparticle comprising: one or more of a man made clathrin light chain protein comprising at least one of a drug activity, and the preceding protein has an amino acid sequence of at least 50, 60, 70, 80, or 90% sequence identity when compared to SEQ ID NO: 12, and administering to a subject in need thereof the preceding composition or protein, wherein the patient is treated for at least one of a disease, condition, or disorder. 
     
     
         2 . The isolated nanoparticle of  claim 1 , optionally comprising, the man made non-cage clathrin light chain protein and bound together with a clathrin heavy chain protein. 
     
     
         3 . The isolated nanoparticle of  claim 1 , wherein the drug is a micromolecule biomolecular drug, macromolecule biomolecular drug, small molecule drug, large molecule drug, or a combination thereof. 
     
     
         4 . The isolated nanoparticle of  claim 1 , wherein the clathrin light chain transports one or more of a drug. 
     
     
         5 . The isolated nanoparticle of  claim 1 , wherein binding one or more of a drug together with the clathrin light comprises a modification of one or more use or effect of the drug. 
     
     
         6 . The isolated nanoparticle of  claim 1 , wherein the isolated nanoparticle is capable of comprising a ligand. 
     
     
         7 . The isolated nanoparticle of  claim 1 , wherein the isolated nanoparticle is capable of comprising a targeting agent. 
     
     
         8 . The isolated nanoparticle of  claim 1 , wherein one or more of a drug is fused together with the clathrin light chain. 
     
     
         9 . The isolated nanoparticle of  claim 1 , wherein one or more of a drug is hybridized with the clathrin chain. 
     
     
         10 . The isolated nanoparticle of  claim 1 , wherein one or more of a drug is bound together with the clathrin light chain by a linker. 
     
     
         11 . The isolated nanoparticle of  claim 10 , wherein the linker is an amino acid, a biotin-avidin coupler, a chemical, a covalent bond, a non-covalent bond, a cross-linker, a molecular bridge, a PEG, a peptide, a molecular bridge, a molecular tether or a spacer or a combination thereof. 
     
     
         12 . The isolated nanoparticle of  claim 1 , further comprising, a masking moiety. 
     
     
         13 . The isolated nanoparticle of  claim 1 , further comprising, a targeting moiety. 
     
     
         14 . The isolated nanoparticle of  claim 1 , wherein the clathrin light chain further comprises, tethers for capturing and positioning one or more elements on the clathrin light chain. 
     
     
         15 . The isolated nanoparticle of  claim 1 , wherein the clathrin light chain is capable of comprising a scaffold for supporting one or more elements. 
     
     
         16 . The isolated nanoparticle of  claim 1 , wherein the clathrin light chain and one or more of a drug is formulated as a pharmaceutical composition. 
     
     
         17 . The isolated nanoparticle of  claim 1 , wherein the pharmaceutical composition optionally comprises, the clathrin light chain and bound together with a clathrin heavy chain. 
     
     
         18 . The isolated nanoparticle of  claim 17 , wherein the clathrin heavy chain further comprises, at least one of a drug activity, 
     
     
         19 . The isolated nanoparticle of  claim 16 , wherein the pharmaceutical composition is formulated for any suitable route and means of administration.

The isolated nanoparticle of  claim 16 , wherein the pharmaceutical composition is formulated as an adhesive, aerosol, biologic, capsule, chemical compound, eye drop, gel, coated, injectable, polymer, liquid, oil, ointment, prosthetic, salve, soft galantine capsule, stent, controlled release, subcutaneous, surgical, syrup, tablet, topical, vapor, or water soluble composition. 
     
     
         21 . The isolated nanoparticle of  claim 1 , wherein the drug activity comprises, internal or external stimuli. 
     
     
         22 . The isolated bio-nanoparticle according to  claim 21 , wherein a stimulus response comprises transport or element release, delivery, activation, assay, sensor, test, reporting, deactivation, therapeutic, diagnostic, self-assembly, disassembly.

The isolated nanoparticle of  claim 21 , wherein the stimulus includes one or more stimuli of any type including but not limited to an acoustical, chemical, biochemical, biological, fluidic, metabolic, covalent, non-covalent, ionic, disorder, disease, electrical, electromagnetic, genotype, heat shock cognate protein, magnetic, mechanical, phenotype, photonic, toxin, temperature, pH, pathogen, pathology, quantum mechanical, radioactive, and radiological or a sonic stimuli type. 
     
     
         24 . The isolated nanoparticle of  claim 1 , wherein the isolated nanoparticle is non-immunogenic and/or non-toxic. 
     
     
         25 . The isolated nanoparticle of  claim 1 , wherein the man-made clathrin light chain protein is stable in serum. 
     
     
         26 . The isolated nanoparticle of  claim 1 , wherein the composition or protein further comprises, molecular tethers that scavenge free radicals or toxins. 
     
     
         27 . The isolated nanoparticle of  claim 1 , wherein the man made clathrin light chain is formed from a plurality of purified, synthetic, recombinant, biologically modified, chemically modified, genetically modified, biologically active, biologically inert, hybridized, chiral molecules, or a combination thereof. 
     
     
         28 . The isolated nanoparticle of  claim 1 , wherein the clathrin light chain comprises a multiple of the man made protein. 
     
     
         29 . The isolated nanoparticle of  claim 1 , wherein SEQ ID NO: 12 is substituted with an alternative light chain protein sequence. 
     
     
         30 . The isolated nanoparticle of  claim 29 , wherein the alternative protein sequence is SEQ ID NO: 13. 
     
     
         31 . The isolated nanoparticle of  claim 1 , comprising, one or more configuration of the man-made clathrin light chain protein. 
     
     
         32 . A method for an isolated nanoparticle comprising: one or more of a man made clathrin light chain protein comprising at least one of a drug activity, and the preceding protein has an amino acid sequence of at least 50, 60, 70, 80, or 90% sequence identity when compared to SEQ ID NO: 12, and administering to a subject in need thereof the preceding composition or protein, wherein the patient is treated for at least one of a disease, condition, or disorder.
