Abstract

A method of correcting singletons in a selected AAV sequence in order to increasing the packaging yield, transduction efficiency, and/or gene transfer efficiency of the selected AAV is provided. This method involves altering one or more singletons in the parental AAV capsid to conform the singleton to the amino acid in the corresponding position(s) of the aligned functional AAV capsid sequences.

1 . A vector comprising an AAV capsid selected from the group consisting of:
 rh20, which has an amino acid sequence of SEQ ID NO:1 or a sequence having at least 97% identity thereto;   rh32/33, which has an amino acid sequence of SEQ ID NO:2 or a sequence having at least 97% identity thereto;   rh39, which has an amino acid sequence of SEQ ID NO:3 or a sequence having at least 97% identity thereto;   rh46, which has an amino acid sequence of SEQ ID NO:4 or a sequence having at least 97% identity thereto;   rh73, which has an amino acid sequence of SEQ ID NO:5 or a sequence having at least 97% identity thereto;   rh74 which has an amino acid sequence of SEQ ID NO:6 or a sequence having at least 97% identity thereto.
