1 . A biotin derivative having structural formula (I): 
       
         
           
           
               
               
           
         
         wherein: 
         X is S or O; 
         R 1  is selected from H and a derivative group; 
         R 2  is selected from H and a derivative group; 
         Y is O or absent; 
         L is absent or is a linker; 
         R 3  is selected from —OR 4 , —COOR 4 , a reactive group, a protein, a peptide, an amino acid, a dextran, a monosaccharide, a disaccharide, a trisaccharide, an oligosaccharide, a polysaccharide, a hormone, a lipopolysaccharide, a nucleotide, an oligonucleotide, a small molecule, a cell, a microplate, and a microparticle; 
         R 4  is selected from H and a derivative group; 
         wherein at least one of R 1  and R 2  is H; 
         wherein if X is O, at least one of R 1  and R 2  is not H, or Y is O;

         wherein:

         R is selected from H and a derivative group;

 a. providing a biotin derivative of  claim 1 , wherein R 3  is a reactive group; and wherein the linker is present and comprises a detectable moiety;   b. providing a sample comprising a protein comprising a complementary group capable of reacting with the reactive group; and   c. contacting the biotin derivative with the sample to provide a labeled protein.   
     
     
         142 . The method of  claim 141 , wherein the reactive group is an alkyne, and the complementary group is an azide.
