Abstract

The invention in suitable embodiments is directed to using purified, non-cage clathrin protein for purposes of assisting in drug discovery and development. In one aspect, a tool composition, formed in whole or in part from isolated, synthetic and or recombinant amino acid residues comprising in whole or in part one or more protein sequence, forms one or more type of tool for carrying out various elements and activities of drug discovery and development.

In view of the foregoing, what is claimed is: 
     
         1 . A medical research tool composition for use in discovering drugs comprising: a man made non-cage clathrin protein and has an amino acid sequence of at least 50, 60, 70, 80, or 90% sequence identity when compared to SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7 SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 10 or SEQ ID NO: 11, and including at least one of a medical research element of one or more type, wherein a formulation comprised of the preceding protein or composition and at least a portion of the medical research element is used to perform discovery of at least one medicinal aspect of a drug or use thereof, and administering the formulation in vitro or in vivo to one or more type of medical research subject. 
     
     
         2 . The medical research tool composition of  claim 1  further comprising, a man made protein that has at least 50, 60, 70, 80, or 90% sequence identity when compared to SEQ ID NO: 12 or SEQ ID NO: 13, and the preceding protein disposed adjacent to the sequence according to  claim 1 , and binding to the sequence. 
     
     
         3 . The medical research tool composition of  claim 1 , wherein the element is a small or large molecule element. 
     
     
         4 . The medical research tool composition of  claim 1 , wherein the element is a biological element. 
     
     
         5 . The medical research tool composition of  claim 1 , wherein the element is a chemical element. 
     
     
         6 . The medical research tool composition of  claim 1 , wherein the element is bound together with the composition. 
     
     
         7 . The medical research tool composition of  claim 1 , wherein the element is incorporated into the composition. 
     
     
         8 . The medical research tool composition of  claim 1  comprising, a multiple of the protein. 
     
     
         9 . The medical research tool composition of  claim 1  comprising, a multiple of the elements. 
     
     
         10 . The medical research tool composition of  claim 1 , wherein the man made protein has a molecular mass of at least about 170 kilodaltons.

The medical research tool composition of  claim 1 , wherein the man made protein comprises a plurality of purified, synthetic, recombinant, biologically modified, chemically modified, genetically modified, biologically active, biologically inert, hybridized or chiral molecules or a combination thereof. 
     
     
         12 . The medical research tool composition of  claim 1 , wherein the man made protein is stable with respect to dissociation. 
     
     
         13 . The medical research tool composition of  claim 1 , wherein the composition is formulated for any suitable route and means of administration. 
     
     
         14 . The medical research tool composition of  claim 13 , wherein the composition is formulated as an adhesive, aerosol, biologic, capsule, chemical compound, eye drop, gel, coated, injectable, polymer, liquid, oil, ointment, prosthetic, salve, soft galantine capsule, stent, controlled release, subcutaneous, surgical, syrup, tablet, topical, vapor, or water soluble composition. 
     
     
         15 . The medical research tool composition of  claim 1 , wherein the protein further comprises, receptors for capturing and positioning the one or more elements on the protein. 
     
     
         16 . The medical research tool composition of  claim 1 , wherein the protein further comprises, a scaffold for supporting the one or more research elements. 
     
     
         17 . The medical research tool composition of  claim 1 , further comprising a therapeutic agent. 
     
     
         18 . The medical research tool composition of  claim 17 , wherein the therapeutic agent is a drug for treating or diagnosing at least one of a disease, condition or disorder. 
     
     
         19 . A method for discovering at least one medicinal aspect of a drug or use thereof, comprising formulating at least a portion of a medical research element with the protein or composition according to  claim 1 . 
     
     
         20 . The method of  claim 19 , comprising researching at least one of medicinal use of the element. 
     
     
         21 . The method of  claim 19 , comprising discovering at least one of efficacious effect of the element. 
     
     
         22 . The method of  claim 19 , comprising discovering at least one of medicinal use of the element.

The method of  claim 37 , comprising administering the element across at least one of blood brain, blood-cerebrospinal, blood vessel, capillary, cell, cell membrane, circumventricular, epithelial, extracellular, fenestrated capillary, fenestrated vessel, intracellular, intramuscular, lipid, lymphatic, nasal, nasal cavity, nasal mucosa, ocular, olfactory epithelium, olfactory nerve, oral, anal, membrane protein, organelle, paracellular, perineural space, plasma membrane, skin, subarachnoid, subcutaneous, tight junction, trigeminal nerve, vomeronasal, vesicle barrier.

A method for a medical research tool composition for use in discovering drugs comprising: forming a man made non-cage clathrin protein and has an amino acid sequence of at least 50, 60, 70, 80, or 90% sequence identity when compared to SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7 SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 10 or SEQ ID NO: 11, and including at least one of a medical research element of one or more type, wherein a formulation comprised of the preceding protein or composition and at least a portion of the medical research element is used to perform discovery of at least one medicinal aspect of a drug or use thereof, and administering the formulation in vitro or in vivo to one or more type of medical research subject.
