Claims
exact text as granted — not AI-modified1 .- 65 . (canceled)
66 . A humanized 1B7 antibody comprising a humanized light chain variable region and a humanized heavy chain variable region,
wherein said humanized light chain variable region comprises:
(i) a mouse CDR L1 as set forth in SEQ ID NO:3, a mouse CDR L2 as set forth in SEQ ID NO:4, a mouse CDR L5 as set forth in SEQ ID NO:5 and
(ii) a tyrosine at a position corresponding to Kabat position 71, a serine at a position corresponding to Kabat position 10, a leucine at a position corresponding to Kabat position 11, a glycine at a position corresponding to Kabat position 16, an arginine at a position corresponding to Kabat position 18, a glycine at a position corresponding to Kabat position 41, a lysine at a position corresponding to Kabat position 42, or a leucine at a position corresponding to Kabat position 47; and
wherein said humanized heavy chain variable region comprises:
(iii) a mouse CDR H1 as set forth in SEQ ID NO:6, a mouse CDR H2 as set forth in SEQ ID NO:7, and a mouse CDR H3 as set forth in SEQ ID NO:8 and
(iv) a valine at a position corresponding to Kabat position 71, an arginine at a position corresponding to Kabat position 38, an alanine at a position corresponding to Kabat position 40, or a valine at a position corresponding to Kabat position 109.
67 . The humanized 1B7 antibody of claim 66 , wherein said humanized light chain variable region comprises a tyrosine at a position corresponding to Kabat position 71 and said humanized heavy chain variable region comprises a valine at a position corresponding to Kabat position 71.
68 . The humanized 1B7 antibody of claim 66 , wherein said antibody is a Fab′ fragment.
69 . The humanized 1B7 antibody of claim 66 , wherein said antibody comprises a human constant region.
70 . The humanized 1B7 antibody of claim 66 , wherein said antibody is an IgG.
71 . The humanized 1B7 antibody of claim 66 , wherein said antibody is a single chain antibody (scFv).
72 . The humanized 1B7 antibody of claim 66 , wherein said antibody is capable of binding a pertussis toxin protein with an equilibrium dissociation constant (K D ) less than 10 nM.
73 . The humanized 1B7 antibody of claim 72 , wherein said pertussis toxin protein is a truncated pertussis toxin protein.
74 . The humanized 1B7 antibody of claim 73 , wherein said truncated pertussis toxin protein comprises the amino acid sequence corresponding to amino acid residues 1 to 220 of SEQ ID NO:9.
75 . The humanized 1B7 antibody of claim 66 , wherein said humanized antibody is an antibody which competes with an antibody that is capable of binding a pertussis toxin protein comprising the amino acid sequence corresponding to amino acid residues 1 to 220 of SEQ ID NO:9.
76 . A humanized antibody which binds the same epitope as the antibody set forth in claim 66 .
77 . A pharmaceutical composition comprising a therapeutically effective amount of a humanized antibody as set forth in claim 66 .
78 . An isolated nucleic acid encoding a humanized antibody as set forth in claim 66 .
79 . A method of treating whooping cough in a subject in need thereof, comprising administering to the subject a therapeutically effective amount of a humanized antibody as set forth in claim 66 and a pharmaceutically acceptable excipient, thereby treating whooping cough in the subject.Join the waitlist — get patent alerts
