Abstract

A preparation comprises a herbal extract and/or fraction, and a pharmaceutical preparation includes extract of Curcuma mangga Val. et Zipp., which has bioactivities in reducing expression levels of 5-alpha-reductase-1, androgen receptor, and PI3 in prostate cancer cells. The use of this present invention is directed to reduce prostate enlargement. Moreover, it also can be used to treat prostate cancer, lung cancer, and other diseases related to GPCR pathway.

1 . An extract of  Curcuma mangga  Val. et. Zipp. and/or its fraction derived therefrom, wherein the extract is obtained by the process comprising the steps of:
 a) Chopping dried  Curcuma mangga  Val. et Zipp. to a length of 3-7 mm;   b) Extracting the chopped material from step (a) with maceration and/or percolation method using organic solvent wherein the solid-to-solvent ratio is (1:6)-(1:10) w/v, the duration of maceration and/or percolation is between 30-240 minutes, and the solids are filtered to obtain filtrate;   c) Concentrating the filtrate from step (b) by evaporation until extract concentrate is obtained;   d) Adding a filler to the extract concentrate from step (c);   e) Drying the mixture of extract concentrate and filler from step (d) to obtain a dry extract;   f) Milling the dry extract from step (e).   
     
     
         2 . The extract and/or fraction obtained through a process according to  claim 1 , wherein the part of  Curcuma mangga  Val. et. Zipp used in step (a) is the rhizome. 
     
     
         3 . The extract and/or fraction obtained through a process according to  claim 1 , wherein the filtrate that is produced in step (b) can be further extracted using liquid-liquid partition method. 
     
     
         4 . The extract and/or fraction obtained through a process according to  claim 1 , wherein the evaporation process of step (c) is performed at a temperature range of 50°-60° C. and at a pressure range of 175-875 mbar. 
     
     
         5 . The extract and/or fraction obtained through a process according to  claim 1 , wherein the drying process of step (e) uses an oven at a temperature range of 60°-70° C. for 24-48 hours. 
     
     
         6 . The extract and/or fraction according to  claim 1 , wherein the extract and/or fraction comprises at least group of compound(s) including flavonoids, saponins, phenols, terpenoids, steroids or its combination. 
     
     
         7 . The extract and/or fraction according to  claim 1 , either as a single active compound or in combination, in an effective amount or dosage as anti-benign prostatic hyperplasia and an inhibitor of prostate cancer cells growth. 
     
     
         8 . The extract and/or fraction according to  claim 7 , wherein the extract and/or fraction reduces the gene expression of 5-alpha-reductase-1. 
     
     
         9 . The extract and/or fraction according to  claim 7 , wherein the extract and/or fraction reduces the gene expression of androgen receptor. 
     
     
         10 . The extract and/or fraction according to  claim 7 , wherein the extract and/or fraction inhibits the PI3 kinase and Akt pathways. 
     
     
         11 . Use of extract and/or fraction according to  claim 1 , as an inhibitor of lung cancer cells growth. 
     
     
         12 . Use of extract and/or fraction according to  claim 1 , as a treatment for disease(s) that related to GPCR pathway, including immune system and hemostatic, cell cycle pathway, biological oxidation, diabetes, cholesterol, as well as lipid and lipoprotein metabolism. 
     
     
         13 . A pharmaceutical composition comprising the extract and/or fraction according to  claim 1 , as a single active ingredient or in combination, and carrier, excipient(s) or additive(s) that are pharmaceutically acceptable and physiologically suitable. 
     
     
         14 . A pharmaceutical composition comprising the extract and/or fraction according to  claim 1 , as a single active ingredient or in combination, and carrier, excipient(s) or additive(s) that are pharmaceutically acceptable and physiologically suitable, wherein the pharmaceutical composition is prepared in the form of solid, semi-solid, or liquid, in sterile or non-sterile product(s). 
     
     
         15 . A food or beverage product directed to promote health benefit comprising the extract and/or fraction according to  claim 1 , as a single active ingredient or in combination.
