Abstract

Provided herein are compositions for the treatment and/or prevention of cardiovascular disease (CVD), and methods of application and use thereof. In particular, the present invention provides treatment and/or prevention of cardiovascular disease with 3,3-dimethyl-1-butanol (DMB) and related compounds, and pharmaceutical formulations thereof. In other embodiments, the present invention provides methods of administering a gut flora targeting antibiotic to a subject prior to a procedure that is associated with a risk of causing thrombosis, heart-attack, and/or platelet hyper-responsiveness.

         19 . A method of preventing thrombosis, heart-attack, and/or reducing platelet hyper-responsiveness in a subject undergoing a procedure comprising:
 administering a gut flora targeting antibiotic to a subject prior to a procedure,   wherein said procedure is associated with a risk of causing thrombosis, heart-attack, and/or platelet hyper-responsiveness, and   wherein said administering is under conditions such that said thrombosis and/or said heart attack is prevented, and/or said platelet hyper-responsiveness is reduced.   
     
     
         20 . The method of  claim 19 , wherein said procedure is an instrumented procedure selected from the group consisting of: dental, surgical, colonoscopy, and vascular stenting. 
     
     
         21 . The method of  claim 19 , wherein said gut flora targeting antibiotic is selected from the group consisting of: ciprofloxin, flagyl (metronidazole), vancomycin, neomycin sulfate, and ampicillin. 
     
     
         22 . The method of  claim 19 , wherein said administering is within 50 hours of said procedure. 
     
     
         23 . The method of  claim 19 , further comprising, prior to said procedure, viewing results of a platelet aggregation assay on a sample taken from said subject after said administering said glut flora targeting antibiotic. 
     
     
         24 . A method for the treatment and/or prevention of cardiovascular disease and/or thrombosis comprising:
 a) identifying a subject as having increased platelet aggregation and/or elevated TMAO levels, and   b) administering to the subject a composition comprising a gut targeting antibiotic.   
     
     
         25 . The method of  claim 24 , wherein said gut flora targeting antibiotic is selected from the group consisting of: ciprofloxin, flagyl (metronidazole), vancomycin, neomycin sulfate, and ampicillin. 
     
     
         26 . The method of  claim 24 , wherein said identifying is identifying said subject as having increased platelet aggregation. 
     
     
         27 . The method of  claim 24 , wherein said identifying is identifying said subject as having elevated TMAO levels. 
     
     
         28 . A method comprising:
 a) performing: i) a platelet aggregation assay on a sample from a subject to determine if said sample shows elevated platelet aggregation compared to normal levels, and/or ii) a performing a TMAO level assay on a sample from a subject to determine if said sample shows elevated TMAO levels compared to normal levels; and   b) recommending, and/or generating a reports that recommends, that said subject receive a therapeutic composition for treating cardiovascular disease or thrombosis, wherein said therapeutic composition comprises a gut targeting antibiotic.   
     
     
         29 . The method of  claim 28 , wherein step b) comprises said generating said report. 
     
     
         30 . The method of  claim 28 , wherein said gut flora targeting antibiotic is selected from the group consisting of: ciprofloxin, flagyl (metronidazole), vancomycin, neomycin sulfate, and ampicillin. 
     
     
         31 . The method of  claim 28 , wherein step a) comprises performing said platelet aggregation assay. 
     
     
         32 . The method of  claim 28 , wherein step a) comprises performing said TMAO level assay.
