Abstract
A particle/cell separation device is described which is particularly adapted for neutrophil depletion from a preparation of whole blood or platelet-rich plasma. Also described are blood and platelet rich plasma compositions produced using the device which are neutrophil-depleted.

18 . A cell separation device for producing neutrophil-depleted platelet-rich plasma with centrifugation, said device comprising:
an inlet portion comprising an inlet port for receiving a sample of whole blood; a separation chamber adapted to separate platelet-rich plasma from other components of the whole blood based on differences in specific gravity; a flow path for the platelet-rich plasma comprising an area of constriction adapted to remove neutrophils from the platelet rich plasma; and an outlet portion comprising a collection port in fluid communication with the separation chamber for removing neutrophil-depleted platelet-rich plasma.
19 . The cell separation device of claim 18 , wherein the area of constriction of the flow path has cross-sectional dimension of 10-100 microns.
20 . The cell separation device of claim 19 , wherein the area of constriction of the flow path has cross-sectional dimension of 10-30 microns.
21 . The cell separation device of claim 18 , wherein the device produces platelet-rich plasma having a platelet concentration of between 2-8 times the platelet concentration in the whole blood.
22 . The cell separation device of claim 18 , wherein the neutrophil concentration in the platelet-rich plasma is less than about 1000/μl.
23 . A kit for treating damaged tissue comprising:
(i) a syringe for withdrawing blood from a patient; (ii) a centrifuge device to obtain neutrophil-depleted platelet-rich plasma; the centrifuge device comprising:
an inlet portion comprising an inlet port for receiving a sample of blood;
a separation chamber adapted to separate platelet-rich plasma from other components of the blood based on differences in specific gravity;
a flow path for the platelet-rich plasma comprising an area of constriction adapted to remove neutrophils from the platelet rich plasma; and
an outlet portion comprising a collection port in fluid communication with the separation chamber for removing neutrophil-depleted platelet-rich plasma; and
(iii) a syringe for subcutaneous delivery of the neutrophil-depleted platelet-rich plasma to the damaged tissue.
24 . The kit of claim 23 , wherein the kit does not contain an activator of the neutrophil-depleted platelet-rich plasma.
25 . The kit claim 24 , wherein the syringe for subcutaneous delivery of the neutrophil-depleted platelet-rich plasma comprises a minimally invasive catheter or needle.
26 . The kit of claim 23 , further comprising an activator of the neutrophil-depleted platelet-rich plasma.
27 . The kit of claim 23 , further comprising a pH adjusting agent for the neutrophil-depleted platelet-rich plasma.
28 . The kit of claim 23 , wherein the cell separation device produces platelet-rich plasma having a platelet concentration of between 2-8 times the platelet concentration in the whole blood.
