Abstract
Novel polypeptide combinations, polynucleotides encoding the polypeptides, and related compositions and methods are disclosed for zcytor17-containing multimeric or heterodimer cytokine receptors that may be used as novel cytokine antagonists, and within methods for detecting ligands that stimulate the proliferation and/or development of hematopoietic, lymphoid and myeloid cells in vitro and in vivo. The present invention also includes methods for producing the multimeric or heterodimeric cytokine receptor, uses therefor and antibodies thereto.

An isolated, humanized, monoclonal antibody which specifically binds to a heterodimeric receptor comprising a first receptor and a second receptor, wherein the first receptor comprises amino acid residues 20-519 of SEQ ID NO:111 or amino acid residues 33-532 of SEQ ID NO:5, and the second receptor comprises amino acid residues 28-739 of SEQ ID NO:7, and wherein the antibody inhibits signal transduction through the heterodimeric receptor induced by a polypeptide comprising amino acid residues 27-164 of SEQ ID NO:2.
2 . The antibody of claim 1 , wherein the antibody specifically binds to the first receptor of the heterodimeric receptor.
3 . The antibody of claim 1 , wherein the first receptor comprises amino acid residues 20-732 of SEQ ID NO:111.
4 . The antibody of claim 1 , wherein the first receptor comprises amino acid residues 33-662 of SEQ ID NO:5.
5 . The antibody of claim 1 , wherein the second receptor comprises amino acid residues 28-979 of SEQ ID NO:7.
6 . The antibody of claim 1 , wherein the antibody comprises a human immunoglobulin constant domain.
7 . A composition comprising the antibody of claim 1 and a pharmaceutically acceptable vehicle.
8 . An isolated polynucleotide encoding the antibody of claim 1 .
9 . An expression vector comprising the following operably linked elements:
a) a transcription promoter; b) a DNA segment comprising the polynucleotide of claim 8 ; and c) a transcription terminator.
10 . A cultured cell comprising the expression vector of claim 9 .
11 . The cell of claim 10 , wherein the cell is a Chinese hamster ovary cell.
12 . A method of producing the antibody of claim 1 comprising:
a) culturing a cell according to claim 10 whereby the antibody is produced; and
b) recovering the antibody.
13 . A method of treating an inflammatory disease in a mammal comprising administering to the mammal a therapeutically effective amount of the antibody of claim 1 , wherein the inflammatory disease is selected from the group consisting of atopic dermatitis, dermatitis, psoriasis, psoriatic arthritis and eczema.
14 . The method of claim 13 , wherein the inflammatory disease is atopic dermatitis.
15 . A method of treating an inflammatory disease in a mammal comprising administering to the mammal a therapeutically effective amount of a composition comprising the antibody of claim 1 and a pharmaceutically acceptable vehicle, wherein the inflammatory disease is selected from the group consisting of atopic dermatitis, dermatitis, psoriasis, psoriatic arthritis and eczema.
16 . The method of claim 15 , wherein the inflammatory disease is atopic dermatitis.
