Abstract

The present disclosure features, inter alia, receptor-targeting reagents (e.g., immunotoxic receptor-targeting reagents), which are useful in, e.g., methods of binding a receptor-targeting reagent to a cell and methods for treating a variety of disorders such as, but not limited to, cancers and inflammatory disorders. Also featured are methods, compositions, and kits useful for selecting an appropriate treatment modality for a subject (e.g., a subject with a cancer or inflammatory disorder) and/or treating a variety of disorders such as cell proliferative disorders.

An in vitro method for killing a cell, the method comprising contacting a cell with an immunotoxic receptor-targeting reagent comprising SEQ ID NO:1, SEQ ID NO:3, SEQ ID NO:18, or SEQ ID NO:19, wherein the cell expresses an epidermal growth factor receptor (EGFR), an interleukin-13 receptor (IL13R), or an interleukin-4 receptor (IL4R) and wherein contacting the cell with the immunotoxic receptor-targeting reagent kills the cell.

An in vivo method for treating a cancer in a subject, the method comprising delivering to a subject having, suspected of having, or at risk of developing, a cancer an immunotoxic receptor-targeting reagent comprising SEQ ID NO:1, SEQ ID NO:3, SEQ ID NO:18, or SEQ ID NO:19, wherein the cancer expresses an epidermal growth factor receptor (EGFR), an interleukin-13 receptor (IL13R), or an interleukin-4 receptor (IL4R).

The method of  claim 91 , wherein the cancer is lung cancer, breast cancer, colon cancer, pancreatic cancer, renal cancer, stomach cancer, liver cancer, bone cancer, hematological cancer, neural tissue cancer, melanoma, thyroid cancer, ovarian cancer, testicular cancer, prostate cancer, cervical cancer, vaginal cancer, or bladder cancer. 
     
     
         93 . The method of  claim 91 , wherein the cancer is a glioblastoma. 
     
     
         94 . The method of  claim 91 , further comprising determining if one or more cells of the cancer express an EGFR, an IL13R, or an IL4R.
