Abstract
Disclosed herein are compositions and methods for modulating the production of a protein in a target cell. The compositions and methods disclosed herein are capable of ameliorating diseases associated with protein or enzyme deficiencies.

A method of treating a subject having a deficiency in a functional polypeptide, comprising administering a composition comprising (a) at least one mRNA at least a portion of which encodes the functional secreted polypeptide; and (b) a transfer vehicle comprising a lipid nanoparticle, wherein following administration of said composition said mRNA is expressed in a target cell to produce said functional secreted polypeptide.

A method of treating a subject having a deficiency in a functional secreted polypeptide, comprising administering a composition comprising (a) at least one mRNA at least a portion of which encodes the functional secreted polypeptide; and (b) a transfer vehicle comprising a lipid nanoparticle, wherein following administration of said composition said mRNA is translated in a target cell to produce the functional polypeptide in said target cell at at least a minimum therapeutic level more than one hour after administration.
