Abstract
Embodiments herein generally relate to methods, compositions and uses of CaMKII inhibitors. Other embodiments relate to methods, compositions and uses of agents that target CaMKII. Yet further embodiments relate to compositions, methods and uses of CaMKIIN-derived molecules and other CaMKII inhibitor molecules that inhibit autonomous CaMKII activity. In accordance with these embodiments, compositions that inhibit autonomous CaMKII activity may be used for treating conditions causing neuronal cell death, for treating cancer or for treating neurodegenerative disorders.

1 . A method for treating a subject having or suspected of developing a condition causing neuronal cell death comprising: administering to a subject in need thereof a therapeutically effective amount of a composition comprising one or more of a peptide consisting of SEQ ID NO:23 and SEQ ID NO:24; and a pharmaceutically acceptable derivative or salt thereof.
2 . The method of claim 1 , further comprising a cell-transfer/penetrating agent associated with the peptide.
3 . The method of claim 1 , wherein the condition causing neuronal cell death comprises stroke, brain ischemia, traumatic brain injury (TBI), or neurodegenerative disorders.
4 . The method of claim 1 , further comprising administering one or more additional treatments for inhibiting neuronal cell death to the subject.
5 . The method of claim 1 , wherein the condition causing neuronal cell death comprises an acute condition.
6 . The method of claim 1 , wherein the condition causing neuronal cell death comprises a chronic condition.
7 . The method of claim 5 , wherein the acute condition comprises stroke or traumatic brain injury.
8 . The method of claim 2 , wherein the cell-transfer/penetrating agent comprises tat, ant, meristyl-group, palmityl-group or combination thereof.
9 . The method of claim 1 , further comprising one or more mutations in the peptide sequence.
10 . The method of claim 8 , wherein one or more amino acids of the peptide are substituted for arginine, alanine, valine, lysine or combination thereof.
11 . A method for treating a subject having or suspected of developing a drug addiction comprising: administering to a subject in need thereof a therapeutically effective amount of a composition comprising one or more of a peptide consisting of SEQ ID NO:23 and SEQ ID NO:24; and a pharmaceutically acceptable derivative or salt thereof.
12 . The method of claim 11 , further comprising a cell-transfer/penetrating agent associated with the peptide.
13 . The method of claim 12 , wherein the cell-transfer/penetrating agent comprises tat, ant, meristyl-group, palmityl-group or combination thereof.
14 . A method for treating a subject having a traumatic brain injury comprising: administering to a subject a therapeutically effective amount of a composition comprising one or more of a peptide consisting of SEQ ID NO:23 and SEQ ID NO:24; and a pharmaceutically acceptable derivative or salt thereof.
15 . The method of claim 14 , further comprising a cell-transfer/penetrating agent associated with the peptide.
16 . The method of claim 17 , wherein the traumatic brain injury comprises a traumatic brain injury due to a stroke, ischemia, Alzheimer's disease, Parkinson's disease, spinal cord injury, or drug addiction.
17 . The method of claim 14 , further comprising administering one or more additional treatments to the subject directed to treating the traumatic brain injury.
18 . A method for treating a subject having cancer comprising, administering to a subject a therapeutically effective amount of a composition comprising one or more of a peptide consisting of SEQ ID NO:23 and SEQ ID NO:24; and a pharmaceutically acceptable derivative or salt thereof to reduce viable cancer cells; and treating the subject.
