Abstract
Disclosed herein are cell cultures and enriched cell populations of endocrine precursor cells, immature pancreatic hormone-expressing cells and mature pancreatic hormone-expressing cells. Also disclosed herein are methods of producing such cell cultures and cell populations.

21 . A population of cells expressing markers characteristic of the pancreatic endoderm lineage, wherein said markers are pancreatic duodenal homeobox 1(PDX1) and NKX6 transcription factor related locus 1 (NKX6.1) or pancreas specific transcription factor 1a(PTF1A).
22 . The cell population of claim 1 , wherein said population is derived from a human pluripotent stem cell.
23 . The cell population of claim 1 , wherein said population is in contact with a gamma secretase inhibitor.
24 . The cell population of claim 1 , wherein said population is in contact with a retinoid.
25 . The cell population of claim 1 , wherein said population is in contact with noggin.
26 . The cell population of claim 1 , wherein said markers are PDX1 and NKX6.1.
27 . The cell population of claim 1 , wherein said markers are PDX1, NKX6.1 and PTF1A.

35 . An in vitro differentiated population of pancreatic endoderm cells obtained from the differentiation of pluripotent stem cells, wherein the population expresses PDX1 and NKX6.1 or PTF1A.
36 . The cell population of claim 35 , wherein said pluripotent stem cell is an embryonic stem cell.
37 . The cell population of claim 35 , wherein said population is in contact with a gamma secretase inhibitor.
