Anit-Human Interleukin-20 Antibodies
Abstract
Anti-human IL20 monoclonal antibodies that can reduce IL20 mediated activation of both IL20R1/IL20R2 and IL22R1/IL20R2 receptor complexes in one or more species, including humans, are described, as well as antigen-binding molecules such as, e.g., antigen-binding antibody fragments, antibody derivatives, and multi-specific molecules designed or derived from such antibodies, and methods or producing such antibodies or other antigen-binding molecules. Such antibodies or other antigen-binding molecules can be used for treating various diseases and disorders, including autoimmune or inflammatory diseases or disorders.

A method of treating an inflammatory or autoimmune disorder comprising administering to a patient in need thereof an isolated anti-human IL20 antibody comprising: a heavy chain CDR1 comprising amino acid sequence corresponding to Kabat residues 31-35 of SEQ ID NO:6; a heavy chain CDR2 comprising amino acid sequence corresponding to Kabat residues 50-65 of SEQ ID NO:6; a heavy chain CDR3 comprising amino acid sequence corresponding to Kabat residues 95-102 of SEQ ID NO:6; a light chain CDR1 comprising amino acid sequence corresponding to Kabat residues 24-34 of SEQ ID NO:9; a light chain CDR2 comprising amino acid sequence corresponding to Kabat residues 50-56 of SEQ ID NO:9; and a light chain CDR3 comprising amino acid sequence corresponding to Kabat residues 89-97 of SEQ ID NO:9.

The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the inflammatory or autoimmune disorder is selected from the group consisting of rheumatoid arthritis, juvenile rheumatoid arthritis, psoriasis, psoriatic arthritis, ankylosing spondylitis, Sjögren's syndrome, multiple sclerosis, inflammatory bowel disease, systemic lupus erythematosus, and lupus nephritis.
3 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the isolated anti-human IL20 antibody comprises a heavy chain variable region comprising SEQ ID NO:6 and a light chain variable region comprising SEQ ID NO:9.
4 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the inflammatory or autoimmune disorder comprises rheumatoid arthritis.
5 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the inflammatory or autoimmune disorder comprises psoriatic arthritis.
6 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the inflammatory or autoimmune disorder comprises inflammatory bowel disease.
7 . The method of treating an inflammatory or autoimmune disorder of claim 6 , wherein inflammatory bowel disease is ulcerative colitis.
8 . The method of treating an inflammatory or autoimmune disorder of claim 6 , wherein inflammatory bowel disease is Crohn's disease.
9 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the inflammatory or autoimmune disorder comprises systemic lupus erythematosus.
10 . The method of treating an inflammatory or autoimmune disorder of claim 1 , wherein the isolated anti-human IL20 antibody is of the IgG4 isotype.
11 . A method of treating an inflammatory or autoimmune disorder comprising administering to a patient in need thereof a pharmaceutical composition comprising:
an isolated anti-human IL20 antibody comprising: a heavy chain CDR1 comprising amino acid sequence corresponding to Kabat residues 31-35 of SEQ ID NO:6; a heavy chain CDR2 comprising amino acid sequence corresponding to Kabat residues 50-65 of SEQ ID NO:6; a heavy chain CDR3 comprising amino acid sequence corresponding to Kabat residues 95-102 of SEQ ID NO:6; a light chain CDR1 comprising amino acid sequence corresponding to Kabat residues 24-34 of SEQ ID NO:9; a light chain CDR2 comprising amino acid sequence corresponding to Kabat residues 50-56 of SEQ ID NO:9; and a light chain CDR3 comprising amino acid sequence corresponding to Kabat residues 89-97 of SEQ ID NO:9; and a pharmaceutically acceptable excipient.
12 . A method of treating an inflammatory or autoimmune disorder of claim 11 , wherein the pharmaceutical composition comprises the isolated anti-human IL20 antibody in a concentration of at least about 80 mg/ml.
13 . The method of treating an inflammatory or autoimmune disorder of claim 11 , wherein the isolated anti-human IL20 antibody comprises a heavy chain variable region comprising SEQ ID NO:6 and a light chain variable region comprising SEQ ID NO:9.
14 . The method of treating an inflammatory or autoimmune disorder of claim 13 , wherein the inflammatory or autoimmune disorder comprises rheumatoid arthritis.
15 . The method of treating an inflammatory or autoimmune disorder of claim 13 , wherein the inflammatory or autoimmune disorder comprises psoriatic arthritis.
16 . The method of treating an inflammatory or autoimmune disorder of claim 13 , wherein the inflammatory or autoimmune disorder comprises inflammatory bowel disease.
17 . The method of treating an inflammatory or autoimmune disorder of claim 16 , wherein inflammatory bowel disease is ulcerative colitis.
18 . The method of treating an inflammatory or autoimmune disorder of claim 16 , wherein inflammatory bowel disease is Crohn's disease.
