Abstract

Compositions and methods for stimulating an immune response against a secreted enterohemorragic Escherichia coli (EHEC) antigen are disclosed. The compositions comprise EHEC cell culture supernatants.

1 . A composition capable of reducing colonization and/or shedding of enterohemorrhagic  Escherichia coli  (EHEC) in a ruminant, the composition comprising an effective amount of an EHEC cell culture supernatant and an immunological adjuvant. 
     
     
         2 . The composition of  claim 1 , the immunological adjuvant is present in the composition at a concentration of about 20% to about 40% (v/v). 
     
     
         3 . The composition of  claim 1 , wherein the immunological adjuvant is present in the composition at a concentration of about 30% (v/v). 
     
     
         4 . The composition of  claim 1 , wherein the immunological adjuvant comprises oil. 
     
     
         5 . The composition of  claim 1 , wherein the immunological adjuvant comprises an oil-in-water emulsion. 
     
     
         6 . The composition of  claim 1 , wherein the immunological adjuvant comprises a mineral oil. 
     
     
         7 . The composition of  claim 1 , wherein the composition comprises an oil-in-water emulsion. 
     
     
         8 . The composition of  claim 1 , wherein the composition further comprises one or more recombinant EHEC antigens selected from the group consisting of EspA, EspB, EspD, Tir, and Intimin. 
     
     
         9 . The composition of  claim 1 , wherein the EHEC is EHEC O157:H7. 
     
     
         10 . The composition of  claim 1 , wherein the composition is capable of reducing colonization of EHEC in a ruminant. 
     
     
         11 . The composition of  claim 1 , wherein the composition is capable of reducing shedding of EHEC in a ruminant. 
     
     
         12 . The composition of  claim 1 , wherein the ruminant is a bovine subject. 
     
     
         13 . The composition of  claim 1 , wherein the EHEC cell culture supernatant is produced by a process comprising culturing EHEC under conditions that favor Type III antigen secretion. 
     
     
         14 . The composition of  claim 13 , wherein the process comprises culturing the EHEC in a cell culture media comprising minimal media supplemented with about 20-100 mM NaHCO 3 . 
     
     
         15 . The composition of  claim 14 , wherein the minimal media is further supplemented with about 5-10 mM MgSO 4 , about 0.1-1.5% glucose, and about 0.05-5% amino acids. 
     
     
         16 . The composition of  claim 13 , wherein the EHEC is cultured in the presence of about 2-10% CO 2  at a temperature of about 37° C. 
     
     
         17 . The composition of  claim 13 , wherein the EHEC is cultured to an optical density (OD) of about 0.7-0.8 at 600 nm. 
     
     
         18 . A method for reducing colonization of EHEC in a ruminant, comprising administering a therapeutically effective amount of the composition of  claim 1 . 
     
     
         19 . A method for reducing shedding of EHEC in a ruminant, comprising administering a therapeutically effective amount of the composition of  claim 1 . 
     
     
         20 . A method for eliciting an immunological response in a mammal against EHEC, comprising administering a therapeutically effective amount of the composition of  claim 1 .
