Abstract

The invention relates to antibody molecules having specificity for antigenic determinants of both IL-17A and IL-17F, therapeutic uses of the antibody molecules and methods for producing said antibody molecules.

         38 . A neutralising antibody which binds human IL-17A and human IL-17F, comprising a heavy chain wherein a variable domain of the heavy chain comprises a sequence given in SEQ ID NO:1 for CDR-H1, a sequence given in SEQ ID NO:2 for CDR-H2 and a sequence given in SEQ ID NO:3 for CDR-H3. 
     
     
         39 . The neutralising antibody which binds human IL-17A and human IL-17F according to  claim 38 , additionally comprising a light chain, wherein a variable domain of the light chain comprises a sequence given in SEQ ID NO:4, for CDR-L1, a sequence given in SEQ ID NO:5 for CDR-L2 and a sequence given in SEQ ID NO:6 for CDR-L3. 
     
     
         40 . A neutralising antibody which binds human IL-17A and human IL-17F, wherein a heavy chain comprises a sequence given in SEQ ID NO:9 or a sequence at least 90% identical thereto. 
     
     
         41 . The neutralising antibody according to  claim 40 , additionally comprising a light chain, wherein the light chain comprises a sequence given in SEQ ID NO:7 or a sequence at least 90% identical thereto. 
     
     
         42 . The neutralising antibody according to  claim 39 , having the heavy chain comprising a sequence given in SEQ ID NO:9 and the light chain comprising the sequence given in SEQ ID NO:7. 
     
     
         43 . A neutralising antibody which binds human IL-17A and human IL-17F, having a heavy chain comprising a sequence given in SEQ ID NO:15 or a sequence at least 95% identical thereto and a light chain comprising a sequence given in SEQ ID NO:11 or a sequence at least 95% identical thereto. 
     
     
         44 . The neutralising antibody which binds human IL-17A and human IL-17F and which binds to a same epitope on human IL-17A and human IL-17F as the antibody of  claim 42 . 
     
     
         45 . The neutralising antibody which binds human IL-17A and human IL-17F which cross-blocks the binding of the antibody of  claim 42  to human IL-17A and/or human IL-17F and/or IL-17A/F heterodimer and/or is cross-blocked from binding human IL-17A and/or human IL-17F and/or IL-17A/F heterodimer by the antibody of  claim 42 . 
     
     
         46 . The neutralising antibody of  claim 38 , wherein the antibody is a whole antibody or a functionally active fragment or derivative thereof. 
     
     
         47 . The neutralising antibody according to  claim 46  where the antibody fragment is a domain antibody, Fab, Fab′, F(ab′) 2 , scFv or an epitope-binding fragment thereof. 
     
     
         48 . The neutralising antibody according to  claim 47 , wherein the antibody or fragment thereof is a CDR-grafted antibody or a fully human antibody. 
     
     
         49 . The antibody of  claim 38 , wherein the antibody or fragment thereof is conjugated to one or more effector molecule(s). 
     
     
         50 . An isolated DNA sequence encoding the heavy and/or light chain(s) of an antibody of  claim 38 . 
     
     
         51 . A cloning or expression vector comprising one or more DNA sequences according to  claim 50 . 
     
     
         52 . The vector according to  claim 51 , wherein the vector comprises a sequence given in SEQ ID NO:18. 
     
     
         53 . The vector according to  claim 52 , wherein the vector comprises a sequences given in SEQ ID NO:14 and the sequence given in SEQ ID NO:18. 
     
     
         54 . A host cell comprising one or more cloning or expression vectors according to  claim 51 . 
     
     
         55 . A process for the production of the antibody of  claim 38 , comprising culturing the host cell of  claim 54  and isolating the antibody. 
     
     
         56 . A pharmaceutical composition comprising an antibody of  claim 38 , in combination with one or more of a pharmaceutically acceptable excipient, diluent or carrier. 
     
     
         57 . The pharmaceutical composition according to  claim 56 , additionally comprising other active ingredients. 
     
     
         58 . The antibody of  claim 38  or a pharmaceutical composition according to  claim 56 , for use in the treatment or prophylaxis of a pathological disorder that is mediated by IL-17A and/or IL-17F or that is associated with an increased level of IL-17A and/or IL-17F.
