Abstract
The present invention provides for the identification, isolation and uses of mammalian Monoamine Oxidase C (MAO-C), also known as renalase.
Claims
exact text as granted — not AI-modified1 - 69 . (canceled)
70 . An isolated renalase protein capable of metabolizing catecholamines, comprising:
an FAD-binding site with bound FAD cofactor and an amine oxidase domain, the renalase protein comprising an amino acid sequence having at least 90% sequence identity to SEQ ID NO:2, wherein the renalase protein is recombinantly produced in a host cell in the presence of exogenous flavin adenine dinucleotide (FAD).
71 . The renalase protein of claim 70 , wherein the renalase protein comprises an amino acid sequence having at least 95% sequence identity to SEQ ID NO:2.
72 . The renalase protein of claim 70 , wherein the renalase protein comprises an amino acid sequence having at least 98% sequence identity to SEQ ID NO:2.
73 . The renalase protein of claim 70 , wherein the renalase protein comprises the amino acid sequence of SEQ ID NO:2.
74 . The renalase protein of claim 70 , wherein the renalase protein comprises an amino acid sequence encoded by SEQ ID NO:1 or SEQ ID NO:3.
75 . The renalase protein of claim 70 , wherein the catecholamine is dopamine, epinephrine, or norepinephrine.
76 . The renalase protein of claim 70 , wherein the host cell is a bacterial host cell.
77 . The renalase protein of claim 70 , wherein the host cell is a yeast cell.
78 . The renalase protein of claim 70 , wherein the host cell is an insect or vertebrate host cell.
79 . The renalase protein of claim 70 , wherein the renalase protein is produced in the presence of exogenous FAD added at the time of inducing protein synthesis.
80 . The renalase protein of claim 70 , wherein the renalase protein comprises a purification tag.
81 . The renalase protein of claim 70 , wherein the renalase protein is produced by denaturing and refolding.
82 . The renalase protein of claim 81 , wherein the renalase protein is produced by recovering from inclusion bodies, denaturing with urea, and refolding.
83 . A pharmaceutical composition comprising the renalase protein of claim 70 and a pharmaceutically acceptable carrier, diluent, or vehicle.
84 . The pharmaceutical composition of claim 83 , formulated for injectable administration.
85 . A method for identifying a subject in need of renalase therapy, comprising:
determining the level of circulating renalase protein in the subject, wherein a subject having lower levels of renalase protein as compared to a normal level is in need of renalase therapy.
86 . The method of claim 85 , wherein the subject has a cardiovascular disease or renal disease.
87 . The method of claim 85 , wherein the level of renalase protein is determined using an antibody specific for renalase.Join the waitlist — get patent alerts
