Glyphosate resistant 5-enolpyruvylshikimate-3-phosphate (epsp) synthase
Abstract
A plant comprising SEQ. ID. NO. 2 or a functional portion thereof, wherein SEQ ID NO. 2 is not native to said plant. A glyphosate resistant grass of economic value comprises a nucleic acid molecule that encodes a EPSPS enzyme. In some embodiments, the nucleic acid molecule comprises a sequence of SEQ. ID. NO. 1, or a functional portion thereof. In some embodiments, the EPSPS enzyme can be a polypeptide molecule comprising an amino acid sequence that is essentially of SEQ. ID. NO. 2, or portion thereof. Embodiments include a DNA construct comprising a SEQ. ID. NO. 1 or a functional portion thereof and transgenic methods for inserting the DNA construct into a plant. Some embodiments include non-transgenic glyphosate resistant turfgrasses.

74 . An isolated glyphosate resistant gene obtained from the germplasm deposited under American Type Cultural Collection as Penner Ryegrass PTA-8157 deposited Jan. 19, 2007.
75 . The isolated glyphosate resistant gene according to claim 74 , wherein the gene comprises a DNA sequence at least 93% homologous to SEQ ID NO:1 that encodes a functional glyphosate-resistant 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS).
76 . The isolated glyphosate resistant gene according to claim 75 , wherein the gene comprises a DNA sequence 100% identical to SEQ ID NO:1.
77 . An isolated nucleic acid molecule obtained from the germplasm deposited under American Type Cultural Collection as Penner Ryegrass PTA-8157 deposited Jan. 19, 2007 comprising SEQ ID NO:1 or a functional portion thereof.
78 . The isolated nucleic acid molecule according to claim 77 , wherein the nucleic acid molecule encodes a glyphosate-resistant 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) having an amino acid sequence at least 80% homologous to SEQ ID NO:2.
79 . The isolated nucleic acid molecule according to claim 78 , wherein the nucleic acid molecule encodes a glyphosate-resistant EPSPS having an amino acid sequence at least 90% homologous to SEQ ID NO:2.
80 . The isolated nucleic acid molecule according to claim 77 , wherein the nucleic acid molecule encodes a glyphosate-resistant EPSPS having an amino acid sequence 100% homologous to SEQ ID NO:2.
81 . An isolated DNA construct comprising the isolated nucleic acid molecule according to claim 77 .
82 . A DNA construct comprising a polynucleotide segment that has a base sequence of SEQ ID NO:1, or a functional portion thereof, and that encodes SEQ ID NO:2, the polynucleotide segment being operably attached to expression regulatory elements that can be used by a host cell to express a 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) polypeptide encoded by the polynucleotide segment.
83 . A recombinant DNA molecule comprising a structural DNA segment that encodes a 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) polypeptide comprising sequence of SEQ ID NO:2, or a functional portion thereof, operably linked to expression regulatory elements that can be used by a host cell to express the EPSPS polypeptide or peptide encoded by the structural DNA segment.
84 . The recombinant DNA molecule according to claim 83 , wherein the expression regulatory elements are those that can be used by a plant cell to achieve expression of the structural DNA sequence.
85 . The recombinant DNA molecule according to claim 83 , wherein the expression regulatory elements include a plant promoter that is heterologous to the structural DNA sequence.
86 . The recombinant DNA molecule according to claim 83 further comprising a DNA sequence that is operably linked to the structural DNA segment and that encodes an amino-terminal chloroplast transit peptide.
87 . The recombinant DNA molecule according to claim 83 , wherein structural DNA segment that encodes the EPSPS polypeptide has a nucleic acid sequence at least 93% identical to SEQ ID NO:1.
88 . An isolated glyphosate-resistant 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) encoded by a DNA molecule comprising SEQ ID NO:1 or a functional portion thereof.
89 . An isolated glyphosate resistant 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) comprising the amino acid sequence of SEQ ID NO:2, or a functional portion SEQ ID NO:2, obtained from the germplasm deposited under American Type Cultural Collection as Penner Ryegrass PTA-8157 deposited Jan. 19, 2007.
