Abstract

Disclosed are specific binding agents, such as fully human antibodies, that bind to angiopoietin-2. Also disclosed are heavy chain fragments, light chain fragments, and CDRs of the antibodies, as well as methods of making and using the antibodies.

We claim: 
     
         1 . An isolated antibody or antigen-binding fragment thereof that specifically binds angiopoietin-2 (Ang-2), wherein the antibody or antigen-binding fragment thereof neutralizes the Ang-2:Tie2 interaction with an IC50 of about 2.81 nM to about 0.08 nM. 
     
     
         2 . The antibody or antigen-binding fragment thereof of  claim 1 , wherein the antibody or antigen-binding fragment thereof neutralizes the Ang-2:Tie2 interaction with an IC50 of about 1.37 nM to about 0.08 nM. 
     
     
         3 . The antibody or antigen-binding fragment thereof of  claim 1 , wherein the antibody or antigen-binding fragment thereof neutralizes the Ang-2:Tie2 interaction with an IC50 of about 0.97 nM to about 0.08 nM. 
     
     
         4 . The antibody or antigen-binding fragment thereof of  claim 1 , wherein the antibody or antigen-binding fragment thereof neutralizes the Ang-2:Tie2 interaction with an IC50 of about 0.56 nM to about 0.08 nM. 
     
     
         5 . The antibody or antigen-binding fragment thereof of  claim 1 , wherein the antibody or antigen-binding fragment thereof neutralizes the Ang-2:Tie2 interaction with an IC50 of about 0.3 nM to about 0.08 nM. 
     
     
         6 . The antibody or antigen-binding fragment thereof of  claim 1 , wherein the IC50 is measured using an ELISA assay. 
     
     
         7 . An isolated antibody or antigen-binding fragment thereof that specifically binds angiopoietin-2 (Ang-2), wherein the antibody or antigen-binding fragment thereof binds Ang-2 with an EC50 of about 0.02 nM to about 0.0005 nM. 
     
     
         8 . The antibody or antigen-binding fragment thereof of  claim 7 , wherein the antibody or antigen-binding fragment thereof binds Ang-2 with an EC50 of about 0.02 nM. 
     
     
         9 . The antibody or antigen-binding fragment thereof of  claim 7 , wherein the antibody or antigen-binding fragment thereof binds Ang-2 with an EC50 of about 0.0005 nM. 
     
     
         10 . The antibody or antigen-binding fragment thereof of  claim 7 , wherein the EC50 is measured using an ELISA assay. 
     
     
         11 . An isolated antibody or antigen-binding fragment thereof that specifically binds angiopoietin-2 (Ang-2), wherein the antibody or antigen-binding fragment thereof binds Ang-2 with a K D  of about 1.2 nM to about 0.12 nM. 
     
     
         12 . The antibody or antigen-binding fragment thereof of  claim 9 , wherein the antibody or antigen-binding fragment thereof binds Ang-2 with a K D  of about 0.12 nM. 
     
     
         13 . The antibody or antigen-binding fragment thereof of  claim 9 , wherein the antibody or antigen-binding fragment thereof binds Ang-2 with a K D  of about 1.2 nM. 
     
     
         14 . The antibody or antigen-binding fragment thereof of  claim 9 , wherein the K D  is calculated using an affinity BIAcore assay. 
     
     
         15 . An isolated antibody or antigen-binding fragment thereof that specifically binds angiopoietin-2 (Ang-2), wherein the antibody or antigen-binding fragment thereof binds amino acids 255-495 of Ang-2. 
     
     
         16 . A pharmaceutical composition comprising the antibody of  claim 1 ,  7 ,  11  or  15  and a pharmaceutically acceptable formulation agent. 
     
     
         17 . A method of treating cancer in a mammal comprising administering a therapeutically effective amount of the antibody or antigen-binding fragment thereof of  claim 1 ,  7 ,  11  or  15 .
