Plant Preservation Systems
Abstract
The invention relates to orthogonal pairs of tRNAs and aminoacyl-tRNA synthetase that can incorporate the unnatural amino acid phenylselenocysteine into proteins produced in eubacterial host cells such as E. coli. The invention provides, for example but not limited to, novel orthogonal aminoacyl-tRNA synthetases, polynucleotides encoding the novel sythetases molecules, methods for identifying and making the novel synthetases, methods for producing containing the unnatural amino acid phenylselenocysteine and translation systems. The invention further provides methods for producing modified proteins (e.g., lapidated proteins) through targeted modification of the phenylselenocysteine residue in a protein.

8 . A flower preservation system comprising:
a sealable container comprising a constriction that defines a stem compartment; a preservative container comprising a preservative liquid; and, one or more cut flowers which comprise one or more blossoms and stems, and which stems comprise cut ends and a plurality of leaves; wherein the flowers are packaged with the cut ends in the preservative container and with at least a portion of a length of the stems and a portion of the leaves in the sealable container; and, wherein the blossoms and a portion of the leaves are substantially outside the stem compartment.
9 . The system of claim 8 , wherein the sealable container or preservative container comprises a plastic film.
10 . The system of claim 8 , wherein the constriction is formed by wrapping a filament around the outside of the sealable container.
11 . The system of claim 10 , wherein the filament is selected from the group consisting of: a rubber band, a wire, a string, a ribbon, tape and a cable tie.
12 . The system of claim 8 , wherein the constriction is formed around the stems at a level along the stems that encloses not more than 80 percent of the leaves in the stem compartment.
13 . The system of claim 8 , wherein the preservative liquid comprises a substantially sugar free preservative media.
14 . The system of claim 8 , wherein the preservative liquid is a preservative media comprising an agar, an auxin, a cytokinin, copper sulfate, citric acid, baking soda and MS salts & vitamins or KM salts & vitamins.
15 . The system of claim 8 , wherein the sealable container further comprises a blossom section.
16 . The system of claim 15 , wherein the blossom section forms a blossom compartment defined at one end by the constriction and is open at the opposite end.
17 . The system of claim 15 , wherein the blossom section comprises a top opening or perforations.
18 . The system of claim 8 , further comprising an outer shipping container or cargo container.
19 . The system of claim 18 , wherein carbon dioxide is supplemented in an environment about the blossoms.

25 . A method of preserving cut flowers, the method comprising:
inserting cut stem ends of one or more flowers into a preservative container of preservative liquid; inserting stems of the one or more flowers into a sealable container, which stems comprise a plurality of leaves; and, constricting the sealable container at a point along the stems, thus forming a stem compartment; wherein a portion of the leaves are enclosed within the stem compartment and at least a portion of the leaves are outside the stem compartment.
26 . The method of claim 25 , wherein said constricting comprises wrapping a filament around the outside of the sealable container.
27 . The method of claim 26 , wherein said constricting further comprises forming a blossom compartment.
28 . The method of claim 27 , further comprising supplementing levels of carbon dioxide in the blossom compartment.
29 . The method of claim 25 , wherein the proportion of the leaves outside the stem compartment is 20 percent or less.
30 . The method of claim 25 , further comprising transporting the flowers in a cargo container by ship or truck.
31 . The method of claim 25 , further comprising controlling a cargo container parameter selected from the group consisting of: temperature, humidity and carbon dioxide level.
32 . The method of claim 25 , further comprising holding the cut flowers in a sealed container without a preservative liquid.
