Abstract

The present disclosure is generally related to methods for combining chemotherapy and immunotherapy for the treatment of a cancer. The methods also relate to generating a drug-resistant cytotoxic immune cell line and uses thereof in conjunction with cytotoxic drugs.

         31 . An isolated composition comprising γδ T-cells genetically engineered to express a polypeptide that confers resistance to a chemotherapy agent. 
     
     
         32 . An isolated composition comprising γδ T-cells comprising a nucleic acid that encodes a polypeptide that confers resistance to a chemotherapy agent.

The composition of  claim 31 , wherein the a polypeptide that confers resistance to a chemotherapy agent is O 6  methylguanine DNA methyltransferase (MGMT), a drug resistant variant of dihydrofolate reductase (L22Y-DHFR), thymidylate synthase, multiple drug resistance-1 protein (MDR1). 
     
     
         34 . An isolated composition comprising natural killer cells wherein greater than about 50% of the natural killer cells express a polypeptide that confers resistance to a chemotherapy agent. 
     
     
         35 . An isolated composition comprising natural killer cells wherein greater than about 50% of the natural killer cells comprise a nucleic acid that encodes a polypeptide that confers resistance to a chemotherapy agent. 
     
     
         36 . An isolated composition consisting essentially of natural killer cells comprising a nucleic acid that encodes a polypeptide that confers resistance to a chemotherapy agent.
