Abstract

GPC (gel permeation chromatography) and size exclusion chromatography with detection by multi-angle light-scattering photometry (SEC-MALS) can be used to accurately measure molecular size and molar mass, respectively, of saccharide conjugates. The invention provides (a) a process for measuring the molecular size of a conjugated saccharide antigen within a sample, comprising a step of analysing the sample by GPC, and (b) a process for measuring the molar mass of a conjugated saccharide antigen within a sample, comprising a step of analysing the sample by SEC-MALS.

1 . A composition comprising a conjugate of a capsular saccharide from serogroup W135  Neisseria meningitidis , wherein the molecular size of the conjugate is 68.7±20% Å and/or the molar mass of the conjugate is 110.1±20% kDa. 
     
     
         2 . The composition of  claim 1  further comprising a conjugate of a capsular saccharide from serogroup A  Neisseria meningitidis.    
     
     
         3 . The composition of  claim 2 , wherein the conjugate of the capsular saccharide from serogroup A  Neisseria meningitidis  has a molecular size of 57.1±20% Å and/or a molar mass of 88.5±20% kDa. 
     
     
         4 . The composition of  claim 1  further comprising a conjugate of a capsular saccharide from serogroup C  Neisseria meningitidis.    
     
     
         5 . The composition of  claim 4 , wherein the conjugate of the capsular saccharide from serogroup C  Neisseria meningitidis  has a molecular size of 57.0±20% Å and/or a molar mass of 85.2±20% kDa. 
     
     
         6 . The composition of  claim 1  further comprising a conjugate of a capsular saccharide from serogroup Y  Neisseria meningitidis.    
     
     
         7 . The composition of  claim 6 , wherein the conjugate of the capsular saccharide from serogroup Y  Neisseria meningitidis  has a molecular size of 63.3±20% Å and/or a molar mass of the conjugate is 84.7±20% kDa.

The composition of  claim 1 , further comprising a conjugate of a capsular saccharide from serogroup A  Neisseria meningitidis  with a molecular size of 57.1±20% Å and/or a molar mass of 88.5±20% kDa; a conjugate of a capsular saccharide from serogroup C  Neisseria meningitidis  with a molecular size of 57.0±20% Å and/or a molar mass of 85.2±20% kDa; and a conjugate of a capsular saccharide from serogroup Y  Neisseria meningitidis  with a molecular size of 63.3±20% Å and/or a molar mass of 84.7±20% kDa. 
     
     
         9 . A composition comprising a conjugate of a capsular saccharide from serogroup Y  Neisseria meningitidis , wherein the molecular size of the conjugate is 63.3±20% Å and/or the molar mass of the conjugate is 84.7±20% kDa. 
     
     
         10 . The composition of  claim 9  further comprising a conjugate of a capsular saccharide from serogroup A  Neisseria meningitidis.    
     
     
         11 . The composition of  claim 10 , wherein the conjugate of the capsular saccharide from serogroup A  Neisseria meningitidis  has a molecular size of 57.1±20% Å and/or the molar mass of the conjugate is 88.5±20% kDa. 
     
     
         12 . The composition of  claim 9  further comprising a conjugate of a capsular saccharide from serogroup C  Neisseria meningitidis.    
     
     
         13 . The composition of  claim 12 , wherein the conjugate of the capsular saccharide from serogroup C  Neisseria meningitidis  has a molecular size of 57.0±20% Å and/or a molar mass of 85.2±20% kDa. 
     
     
         14 . The composition of  claim 9  further comprising a conjugate of a capsular saccharide from serogroup W135  Neisseria meningitidis.    
     
     
         15 . The composition of  claim 14 , wherein the conjugate of the capsular saccharide from serogroup W135  Neisseria meningitidis  has a molecular size of 68.7±20% Å and/or a molar mass of 110.1±20% kDa.

A composition comprising conjugates of capsular saccharides from serogroups A, C, W135, and Y  Neisseria meningitidis , wherein the conjugate of the capsular saccharide from serogroup C  Neisseria meningitidis  has a molecular size of 57.0±20% Å and/or a molar mass of 85.2±20% kDa. 
     
     
         17 . The composition of  claim 16 , wherein the conjugate of the capsular saccharide from serogroup A  Neisseria meningitidis  has a molecular size of 57.1±20% Å and/or the molar mass of the conjugate is 88.5±20% kDa. 
     
     
         18 . The composition of  claim 16 , wherein the conjugate of the capsular saccharide from serogroup W135  Neisseria meningitidis  has a molecular size of 68.7±20% Å and/or a molar mass of 110.1±20% kDa. 
     
     
         19 . The composition of  claim 16 , wherein the conjugate of the capsular saccharide from serogroup Y  Neisseria meningitidis  has a molecular size of 63.3±20% Å and/or a molar mass of 84.7±20% kDa. 
     
     
         20 . A process for measuring the molecular size of a conjugate of a capsular saccharide from  Neisseria meningitidis  within a sample, comprising a step of size exclusion chromatography with detection by multi-angle light-scattering photometry.
