Abstract

The invention discloses a human cancer-related gene, LAPTM4B, its encoded products and their applications thereof. This human cancer-related gene provided by this invention comprises one of the following nucleotide sequences: (1) SEQ ID No: 1, SEQ ID No: 2, SEQ ID No: 3, SEQ ID No: 6, or SEQ ID No: 8 in the sequence listings; (2) SEQ ID No: 4, SEQ ID No: 5, or SEQ ID No: 7 in the sequence listings which are proteins encoded by the polynucleotides in sequences of SEQ ID No: 1, SEQ ID No: 2, SEQ ID No: 3 and SEQ ID No: 6. (3) Applications of the polynucleotides and polypeptides in the listings in diagnosis and treatments of cancer. This invention enables the developments of new cancer diagnostic markers and new anti-cancer medicines. It would create a significant impact on human society.

1 . A method of cancer treatment based on the principle of LAPTM4B being related to cancer occurrence, comprising a first step of regulating a functionality of a protein encoded by a coding region of LAPTM4B. 
     
     
         2 . The method of  claim 1 , wherein said first step is a step of suppressing expression of a coding region of LAPTM4B and said coding region is for encoding SEQ ID NO: 4 or SEQ ID NO: 7. 
     
     
         3 . The method of  claim 1 , wherein said first step is a step of promoting expression of a coding region of LAPTM4B and said coding region is for encoding protein SEQ ID NO: 5. 
     
     
         4 . The method of  claim 2 , wherein said step of suppressing expression is performed by inhibiting translation of said coding region of LAPTM4B into a corresponding protein. 
     
     
         5 . The method of  claim 4 , wherein siRNA is used for interfering translation of said coding region of LAPTM4B. 
     
     
         6 . The method of  claim 1 , wherein said first step is a step of forming an antigen-antibody complex with said protein, which is SEQ ID NO: 4 or SEQ ID NO: 7. 
     
     
         7 . The method of  claim 1 , wherein said first step was performed by inhibiting phosphorylation of a tyrosine residue at position 285 of SEQ ID NO: 4 to inhibit signaling activation for LAPTM4B overexpression. 
     
     
         8 . A method of cancer diagnosis based on the principle of LAPTM4B being related to cancer occurrence, comprising a step of determining the level of expression of a coding region of LAPTM4B. 
     
     
         9 . The method of  claim 8 , wherein said step of determining the level of expression is performed by a method of measuring the amount of mRNA transcribed from LAPTM4B defined by SEQ ID NO: 1 or SEQ ID NO: 6. 
     
     
         10 . The method of  claim 8 , wherein said mRNA encoding for protein SEQ ID. NO: 4. 
     
     
         11 . The method of  claim 8 , wherein said mRNA encoding for protein SEQ ID. NO: 5. 
     
     
         12 . The method of  claim 8 , wherein said mRNA encoding for protein SEQ ID. NO: 7. 
     
     
         13 . The method of  claim 8 , wherein said step of determining the level of expression is performed by a method of measuring the amount of a protein encoded by LAPTM4B defined by SEQ ID NO: 1 or SEQ ID NO: 6 in a specimen from a patient, said protein encoded by LAPTM4B being selected from the group consisting of SEQ ID. NO: 4, SEQ ID. NO: 5. and SEQ ID. NO: 7. 
     
     
         14 . The method of  claim 13 , wherein said specimen is a tissue, a fluid sample or a blood sample from a patient. 
     
     
         15 . The method of  claim 13 , wherein said patient is a subject suffering from liver cancer, lung cancer, stomach cancer, colon cancer and breast cancer. 
     
     
         16 . The method of  claim 13 , wherein said step of determining the level of expression is performed with an antibody based kit. 
     
     
         17 . An antibody, which is useful based on the principle of LAPTM4B being related to cancer occurrence and capable of forming an antibody-antigen complex with a protein encoded by LAPTM4B. 
     
     
         18 . The antibody of  claim 17 , wherein said antibody is directed to protein SEQ ID NO:4 encoded by LAPTM4B polynucleotide SEQ ID. NO: 1. 
     
     
         19 . The antibody of  claim 17 , wherein said antibody is directed to protein SEQ ID NO:7 encoded by LAPTM4B polynucleotide SEQ ID. NO: 6.
