Abstract

Photochemical tissue bonding methods include the application of a photosensitizer to a tissue, e.g., cornea, followed by irradiation with electromagnetic energy to produce a tissue seal. The methods are useful for wound repair, or other tissue repair.

1 . A kit for repairing a corneal lesion comprising:
 a photosensitizer agent; and   instructions for repairing a corneal lesion, wherein said kit excludes an exogenous source of cross-linkable substrate.   
     
     
         2 . The kit of  claim 1 , wherein the photosensitizer agent is selected from the group consisting of Rose Bengal, riboflavin-5-phosphate, and a thiazine dye. 
     
     
         3 . The kit of  claim 1 , wherein the photosensitizer agent is Rose Bengal. 
     
     
         4 . The kit according to  claim 1 , wherein the corneal lesion is caused by a surgical procedure. 
     
     
         5 . The kit of  claim 4 , wherein the surgical procedure is selected from the group consisting of corneal transplant surgery, cataract surgery, laser surgery, keratoplasty, refractive surgery, cornea reshaping, and treatment of corneal laceration. 
     
     
         6 . The kit of  claim 1 , wherein the instructions comprise the steps of:
 contacting a lesion in corneal tissue with at least one photosensitizer agent to form a corneal tissue-photosensitizer mixture; and
 applying electromagnetic energy to the corneal tissue-photosensitizer mixture in a manner effective to elicit the production of a reactive species from the photosensitizer without producing substantial thermal damage, 
 wherein the corneal tissue is not contacted with an exogenous cross-linkable substrate which is cross-linked by the application of electromagnetic energy, 
   thereby promoting a partial or complete repair of the corneal lesion.   
     
     
         7 . The kit of  claim 6 , wherein the electromagnetic energy applied is about 124 to about 762 J/cm 2 . 
     
     
         8 . The kit of  claim 6 , wherein the electromagnetic energy is applied for at least five minutes. 
     
     
         9 . The kit of  claim 6 , wherein electromagnetic energy has a wavelength of at least 488 nm. 
     
     
         10 . The kit of  claim 6 , wherein electromagnetic energy is at an irradiance of less than 3.5 W/cm 2 . 
     
     
         11 . The kit of  claim 6 , wherein the tissue seal that is created has a tensile strength. 
     
     
         12 . The kit of  claim 6 , wherein the partial or complete repair of the corneal lesion is achieved such that the intraocular pressure limit of the repaired corneal lesion is greater than 100 mm Hg. 
     
     
         13 . A kit for repairing a corneal lesion in a living subject, comprising:
 a photosensitizer agent; and   instructions for repairing a corneal lesion, wherein said kit excludes an exogenous source of cross-linkable substrate.   
     
     
         14 . The kit of  claim 13 , wherein the subject is a human. 
     
     
         15 . The kit of  claim 13 , wherein the photosensitizer agent is selected from the group consisting of Rose Bengal, riboflavin-5-phosphate, and a thiazine dye. 
     
     
         16 . The kit of  claim 13 , wherein the photosensitizer agent is Rose Bengal. 
     
     
         17 . The method of  claim 13 , wherein the corneal lesion is caused by a surgical procedure. 
     
     
         18 . The kit of  claim 17 , wherein the surgical procedure is selected from the group consisting of corneal transplant surgery, cataract surgery, laser surgery, keratoplasty, refractive surgery, cornea reshaping, and treatment of corneal laceration. 
     
     
         19 . The kit of  claim 13 , wherein the instructions comprise the steps of:
 contacting a lesion in corneal tissue with Rose Bengal (RB) to form a corneal tissue-RB mixture; and   applying electromagnetic energy to the corneal tissue-RB mixture in a manner effective to elicit the production of a reactive species from the RB without producing substantial thermal damage,   wherein the corneal tissue is not contacted with an exogenous cross-linkable substrate which is cross-linked by the application of electromagnetic energy,   
       thereby promoting a partial or complete repair of the corneal lesion. 
     
     
         20 . The kit of  claim 19 , wherein electromagnetic energy is at an irradiance of less than 3.5 W/cm 2 . 
     
     
         21 . The kit of  claim 19 , wherein the partial or complete repair of the corneal lesion is achieved such that the intraocular pressure limit of the repaired corneal lesion is greater than 100 mm Hg. 
     
     
         22 . The kit of  claim 19 , wherein the electromagnetic energy applied is about 124 to about 762 J/cm 2 .

 a photosensitizer agent; and   instructions for repairing a tissue, wherein said kit excludes an exogenous source of cross-linkable substrate.   
     
     
         27 . The kit of  claim 26 , wherein the tissue is corneal tissue. 
     
     
         28 . The kit of  claim 26 , wherein at least one photosensitizer agent is selected from the group consisting of Rose Bengal, riboflavin-5-phosphate, and a thiazine dye. 
     
     
         29 . The kit of  claim 26 , wherein at least one photosensitizer agent is Rose Bengal. 
     
     
         30 . The kit of  claim 26 , wherein the instructions comprise the steps of:
 contacting a tissue in need of repair, and optionally a second tissue, with at least one photosensitizer agent to form a tissue-photosensitizer mixture; and   applying electromagnetic energy to the tissue-photosensitizer mixture in a manner effective to produce cross linking of a protein in the tissue without producing substantial thermal damage,   wherein the tissue is not contacted with an exogenous cross-linkable substrate which is cross linked by the application of electromagnetic energy,   
       thereby creating a tissue seal that repairs the tissue. 
     
     
         31 . The kit of  claim 30 , wherein electromagnetic energy is at an irradiance of less than 3.5 W/cm 2 . 
     
     
         32 . The kit of  claim 30 , wherein the partial or complete repair of the corneal lesion is achieved such that the intraocular pressure limit of the repaired corneal lesion is greater than 100 mm Hg.
