Abstract

The present invention relates to novel human secreted proteins and isolated nucleic acids containing the coding regions of the genes encoding such proteins. Also provided are vectors, host cells, antibodies, and recombinant methods for producing human secreted proteins. The invention further relates to diagnostic and therapeutic methods useful for diagnosing and treating disorders related to these novel human secreted proteins.

1 . An isolated nucleic acid molecule, comprising a HEMCM42 polynucleotide selected from the group consisting of:
 (a) a polynucleotide encoding amino acid residues 1 to 113 of SEQ ID NO:59;   (b) a polynucleotide encoding amino acid residues 2 to 113 of SEQ ID NO:59;   (c) a polynucleotide encoding amino acid residues 30 to 113 of SEQ ID NO:59;   (d) a polynucleotide encoding the amino acid sequence of the full-length HEMCM42 polypeptide, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No. 209075;   (e) a polynucleotide encoding the amino acid sequence of the HEMCM42 polypeptide, excluding the N-terminal methionine residue, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No. 209075;   (f) a polynucleotide encoding the amino acid sequence of the HEMCM42 polypeptide, excluding the leader sequence of the polypeptide, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No. 209075;   (g) a polynucleotide comprising a first polynucleotide 90% or more identical to a second polynucleotide, wherein the second polynucleotide is the polynucleotide of (a), (b), (c), (d), (e), or (f);   (h) a polynucleotide encoding at least 30 contiguous amino acids of SEQ ID NO:59;   (i) a polynucleotide encoding at least 50 contiguous amino acids of SEQ ID NO:59; and   (j) a polynucleotide comprising a polynucleotide which hybridizes to the complement of any one of the polynucleotides specified in (a) to (i), wherein the hybridization occurs under stringent conditions.   
     
     
         2 . A vector comprising the isolated nucleic acid molecule of  claim 1 . 
     
     
         3 . A method of making an isolated host cell comprising introducing the isolated nucleic acid molecule of  claim 1  into the host cell. 
     
     
         4 . The isolated host cell produced by the method of  claim 3 . 
     
     
         5 . A method of diagnosing a pathological condition or a susceptibility to a pathological condition in a subject comprising:
 (a) determining the presence or absence of a mutation in nucleic acid in a biological sample obtained from the subject when comparing the obtained nucleic acid to the isolated nucleic acid of  claim 1 ; and   (b) diagnosing a pathological condition or a susceptibility to a pathological condition based on the presence or absence of the mutation.   
     
     
         6 . An isolated polypeptide comprising a HEMCM42 polypeptide selected from the group consisting of:
 (a) a polypeptide comprising amino acid residues 1 to 113 of SEQ ID NO:59;   (b) a polypeptide comprising amino acid residues 2 to 113 of SEQ ID NO:59;   (c) a polypeptide comprising amino acid residues 30 to 113 of SEQ ID NO:59;   (d) a polypeptide comprising the amino acid sequence of the full-length HEMCM42 polypeptide, which amino acid sequence is encoded by the cDNA contained in ATCC Deposit No:209075;   (e) a polypeptide comprising the amino acid sequence of the HEMCM42 polypeptide, excluding the N-terminal methionine residue, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No:209075; and   a polypeptide comprising the amino acid sequence of the HEMCM42 polypeptide, excluding the leader sequence of the polypeptide, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No:209075;   (g) a polypeptide comprising a first polypeptide 90% or more identical to a second polypeptide, wherein the second polypeptide is the polypeptide of (a), (b), (c), (d), (e), or (f);   (h) a polypeptide comprising at least 30 contiguous amino acids of SEQ ID NO:59; and   (i) a polypeptide comprising at least 50 contiguous amino acids of SEQ ID NO:59.   
     
     
         7 . A vector comprising an isolated nucleic acid that encodes the isolated polypeptide of  claim 6 . 
     
     
         8 . An isolated host cell that expresses the isolated polypeptide of  claim 6 . 
     
     
         9 . A method of making an isolated polypeptide comprising:
 (a) culturing the isolated host cell of  claim 8  under conditions such that the polypeptide is expressed; and   (b) recovering the polypeptide.   
     
     
         10 . A method for preventing, treating, or ameliorating a medical condition, comprising administering to a mammalian subject a therapeutically effective amount of the polypeptide of  claim 6 . 
     
     
         11 . A method for identifying a binding partner to the polypeptide of  claim 6  comprising:
 (a) contacting the polypeptide of  claim 6  with a potential binding partner; and 
 (b) determining whether the potential binding partner affects an activity of the polypeptide. 
 
     
     
         12 . An isolated antibody or fragment thereof that binds specifically to a HEMCM42 polypeptide selected from the group consisting of:
 (a) a polypeptide comprising amino acid residues 1 to 113 of SEQ ID NO:59;   (b) a polypeptide comprising amino acid residues 2 to 113 of SEQ ID NO:59;   (c) a polypeptide comprising amino acid residues 30 to 113 of SEQ ID NO:59;   (d) a polypeptide comprising the amino acid sequence of the full-length HEMCM42 polypeptide, which amino acid sequence is encoded by the cDNA contained in ATCC Deposit No:209075;   (e) a polypeptide comprising the amino acid sequence of the HEMCM42 polypeptide, excluding the N-terminal methionine residue, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No:209075;   (f) a polypeptide comprising the amino acid sequence of the HEMCM42 polypeptide, excluding the leader sequence of the polypeptide, which amino acid sequence is encoded by a polynucleotide contained within the cDNA contained in ATCC Deposit No:209075;   (g) a polypeptide comprising a first polypeptide 90% or more identical to a second polypeptide, wherein the second polypeptide is the polypeptide of (a), (b), (c), (d), (e), or (f);   (h) a polypeptide comprising at least 30 contiguous amino acids of SEQ ID NO:59; and   (i) a polypeptide comprising at least 50 contiguous amino acids of SEQ ID NO:59.   
     
     
         13 . The isolated antibody or fragment thereof of  claim 12 , wherein the HEMCM42 polypeptide specifically bound by the antibody or fragment thereof is glycosylated. 
     
     
         14 . The isolated antibody or fragment thereof of  claim 12  which is selected from the group consisting of:
 (a) a monoclonal antibody; 
 (b) a polyclonal antibody; 
 (c) a chimeric antibody; 
 (d) a humanized antibody; 
 (e) a single chain antibody; and 
 (f) a Fab fragment. 
 
     
     
         15 . The isolated antibody or fragment thereof of  claim 12  which is labeled. 
     
     
         16 . The isolated antibody or fragment thereof of  claim 12  wherein the isolated antibody or fragment thereof specifically binds to the HEMCM42 polypeptide in a Western blot. 
     
     
         17 . The isolated antibody or fragment thereof of  claim 12  wherein the isolated antibody or fragment thereof specifically binds to the HEMCM42 polypeptide in an ELISA. 
     
     
         18 . An isolated host cell that produces the isolated antibody of  claim 12 . 
     
     
         19 . A hybridoma that produces the isolated antibody of  claim 12 .
